bêta-amyloïde (Aß) est un peptide de 36 à 43 acides aminés qui est formé après clivage séquentiel de la protéine précurseur de l&#39;amyloïde (APP) par β et γ d&#39;une secrétases. La γ-sécrétase traite l&#39;extrémité C-terminale du peptide Aß et est responsable de ses longueurs variables 5. Les formes les plus courantes de Aß sont Aß 40 et Aß 42, ce dernier étant le plus souvent associée à des états pathologiques tels que AD 5. A des concentrations élevées Aß forme β-feuilles qui s&#39;agrègent pour former des fibrilles amyloïdes 6. Fibrilles dépôts sont la principale composante des plaques séniles autour des neurones. Les deux plaques et diffusables, les oligomères de Aß non-plaque, sont pensés pour constituer le sous-jacent pathogènes de Aß. Etude en laboratoire des amylopathies neuronales est compliquée par le fait que ces conditions progrès avec le temps. Par conséquent, il est important de développer animaux génétiquement traitable modèles-complémentaire à des souris-avec durée de vie courte. Ces modèles peuvent être utilisés pour élucider les aspects spécifiques de amylopathies-général cellulaire et moléculaire et en raison de leur simplicité, de les aider à capturer l&#39;essence du problème. Le ver Caenorhabditis elegans chutes est cette catégorie. Il a une durée de vie courte, ~ 20 jours et dans les processus cellulaires de base supplémentaires, y compris la régulation de l&#39;expression génique, le trafic des protéines, de la connectivité neuronale, synaptogenèse, la signalisation cellulaire et la mort sont similaires à des mammifères 7. Amylopathy est un terme qui décrit la synthèse anormale et l&#39;accumulation de bêta-amyloïde (Aß) dans les tissus avec le temps. Aß est une caractéristique de la maladie d&#39;Alzheimer (MA) et se trouve dans la démence à corps de Lewy, la myosite à inclusions et l&#39;angiopathie amyloïde cérébrale 1-4. Amylopathies développer progressivement avec le temps. Pour cette raison, les organismes simples avec une durée de vie courte peuvent aider à élucider les aspects moléculaires de ces conditions. Ici, nous décrivons les protocoles expérimentaux pour étudier la neurodégénérescence Aß induite par l&#39;utilisation des Caenorhabditis elegans du ver. Ainsi, nous construisons vers transgéniques par injection d&#39;ADN codant pour Aß humain 42 dans les gonades syncytial des hermaphrodites adultes. Des lignées transformantes sont stabilisés par une intégration mutagenèse induite. nématodes âge synchronisée par la collecte et l&#39;ensemencement de leurs oeufs. La fonction de neurones exprimant Aß 42 est testé dans des essais comportementaux opportuns (essai de chimiotaxies). Cultures primaires de neurones obtenus à partir d&#39;embryons sont utilisés pour compléter les données comportementales et de tester les effets neuroprotecteurs de composés anti-apoptotiques.

Avec nos protocoles, nous étudions les effets de l&#39;homme Aß 42 oligomères sur la fonction neuronale 8. Un fragment codant Aß humain 42 et le signal de séquence codant pour un peptide artificiel de vecteur d&#39;incendie pPD50.52 été amplifié à partir d&#39;assemblage PCL12 9 en utilisant des amorces qui introduisent un site d&#39;une endonucléase de restriction Sma aux extrémités.
