Het is een uitdaging om een stabiele, inheemse recombinant Mgm101 eiwit uit E. coli mogelijk vanwege zijn onoplosbaarheid in bacteriële cellen. In dit verslag tonen we dat het MBP-fusie-strategie kan het eiwit op een redelijk hoog niveau tot expressie. Door negatieve kleuring transmissie-elektronenmicroscopie en gelpermeatiechromatografie, hebben we eerder aangetoond dat de MBP-fusie-eiwit vormt oligomeren uniform in vitro 18. Homologe recombinatie is belangrijk voor de reparatie van dubbelstrengige breuken (DSB) en InterStrand verknopingen en de her-initiatie van DNA replicatie van ingestorte replicatievorken 1. In conventionele homologe recombinatie, wordt de centrale reactie gekatalyseerd door de ATP-afhankelijke recombinases zoals RecA in prokaryoten en Rad51 en DMC1 in eukaryoten 1-3. Deze recombinasen vormen nucleoproteïne filamenten ssDNA, die essentieel zijn voor het initiëren homologieonderzoek en streng invasie in duplex DNA-matrijzen (figuur 1, linker paneel) 4-7 zijn. Naast de gebruikelijke regeling kan homologe recombinatie ook plaatsvinden in een RecA/Rad51-independent wijze (Figuur 1, rechter paneel). Zo kan de gist Rad59 eiwitten RAD52 en direct katalyseren het hybridiseren van complementaire ssDNA strengen die worden blootgesteld door wegsnijden van dsDNA onderbrekingen. Deze recombinatie proces, bekend als zingenle streng gloeien, over het algemeen niet gepaard homoloog koppeling met dsDNA templates. Na hybridisatie worden heterologe staarten verwijderd door exonucleasen en nicks worden geligeerd aan genoom continuïteit 8-10 herstellen. Reparatie door de enkele streng gloeien mechanisme wordt vaak vergezeld door deleties van genomische sequenties tussen direct herhaalde regio. Rad52 behoort tot een diverse groep van recombinatie eiwitten die wijdverbreid onder bacteriofagen 11. Deze eiwitten zijn ook bekend als Single Strand Gloeien Proteins (SSAPs), op basis van hun activiteit bij het bevorderen van de annealing van de homologe enkelstrengs DNA-moleculen. De beste gekenmerkt bacteriofaag SSAPs zijn Redp en Erf uit de bacteriofagen λ en P22, RecT uit de profaag rac en de Sak eiwit uit de lactococcale faag ul36. De SSAPs structureel zijn typische β-β-β-α voudig, hoewel overeenstemming vrijwel undetectkunnen in de primaire sequenties. Ze maken allemaal grote homo-oligomere ringen van 10 - 14 voudige symmetrie in vitro 12-14. De functionele implicaties van deze karakteristieke hogere orde structurele organisatie is niet goed begrepen. Wij zijn geïnteresseerd in het begrijpen van het mechanisme van homologe recombinatie in het mitochondriaal genoom. We hebben eerder geïdentificeerd MGM101 het gen dat essentieel is voor het behoud van mtDNA in Saccharomyces cerevisiae 15. MGM101 bleek vervolgens worden geassocieerd met mitochondriale nucleoids en is vereist voor de tolerantie van mtDNA aan DNA-beschadigende middelen 16. Echter, de studie van Mgm101 werd weer gehouden in de afgelopen tien jaar door de moeilijkheid om recombinant Mgm101 produceren. We hebben onlangs in geslaagd in het produceren van oplosbare Mgm101 op grote hoeveelheden van E. coli met behulp van de MBP-fusie strategie. Dit heeft ons in staat aan te tonen dat Mgm101 aandelenbiochemische en structurele overeenkomsten met de rad52-familie van eiwitten 17,18. In dit rapport wordt een drie-stappen zuivering procedure beschreven, die homogeen Mgm101for biochemische en structurele analyses (figuur 2) produceert.

De MGM101 gen werd 20 jaar geleden geïdentificeerd voor zijn rol in het onderhoud van mitochondriaal DNA. Studies uit verschillende groepen hebben gesuggereerd dat de Mgm101 eiwit betrokken is bij de reparatie van recombinational mitochondriaal DNA. Recente onderzoekingen hebben aangegeven dat Mgm101 is gerelateerd aan het RAD52 type recombinatie eiwitfamilie. Deze eiwitten vormen grote oligomere ringen en bevordering van de annealing van de homologe enkelstrengs DNA-moleculen. Echter, de karakterisering van Mgm101 gehinderd door de moeilijkheid in het produceren van het recombinante eiwit. Hier is een betrouwbare werkwijze voor de bereiding van recombinant Mgm101 beschreven. Maltose bindend eiwit (MBP)-gelabeld Mgm101 eerst tot expressie in Escherichia coli. Het fusie-eiwit wordt eerst gezuiverd door affiniteitschromatografie amylose. Na zijn vrijlating door proteolytische splitsing, wordt Mgm101 gescheiden van MBP door kationische uitwisselingschromatografie. Monodisperse Mgm101 wordt dan verkregendoor grootte-uitsluitingschromatografie. Een opbrengst van -0,87 mg Mgm101 per liter bacteriekweek kan routinematig verkregen. De recombinant Mgm101 heeft minimale verontreiniging van DNA. De geprepareerde monsters worden met succes gebruikt voor biochemische, structurele en single particle image analyses van Mgm101. Dit protocol kan ook gebruikt worden voor de bereiding van andere grote oligomere DNA-bindende eiwitten die kunnen worden verkeerd gevouwen en giftig voor bacteriële cellen.

Mgm101 een RAD52-gerelateerde recombinatie eiwit in mitochondria. Rad52 is uitgebreid bestudeerd om zijn rol in het mitochondriaal DNA recombinatie (Figuur 1). Recombinant Mgm101 kunnen worden bereid door een drie-stappen-procedure (figuur 2).
