BiFC is een eenvoudige en gevoelige methode om eiwit-eiwit interacties te bestuderen. Deze werkwijze kan niet worden gebruikt om nieuwe eiwit-eiwit interacties te identificeren, maar het is vooral handig om eiwit-eiwit interacties binnen cellen bevestigen en functionele eigenschappen te bestuderen, zoals subcellulaire lokalisatie van eiwitcomplexen, in kaart brengen van eiwit-eiwit interactieplaatsen en voor het screenen van kleine moleculen / peptiden die eiwit-eiwit interacties kunnen moduleren. 650,000 eiwit-eiwit interacties worden geraamd, dat in het menselijk interactome, spelen cruciale rol in het handhaven van normale celfuncties 1,2. Naast co-immunoprecipitatie (co-IP), de gouden standaard eiwit-eiwit interacties te bestuderen van cellysaat, verschillende eiwitbanden fragment complementatie bepalingen (PCA) zijn ontwikkeld om de gevoeligheid in het detecteren van eiwit-eiwit interacties binnen cellen 3 verbeteren. Technieken omvatten Förster-energieoverdracht (FRET), Bioluminescence Resonance Energy Transfer (BRET), en Bimoleculaire Fluorescentie Complementatie (BiFC) 4,5. BiFC is gebaseerd op het vergemakkelijkt associatie van twee fragmenten van een fluorescerend eiwit (hier gebruiken we Venus, een YFP variant) die elk worden gefuseerd aan een potentiële interactie eiwit partner (in dit voorbeeld gebruiken we AKAP LBC-en PDE4D3). Interactie van de twee eiwitten van belang in cellen resulteert in functionele fluorescentie, die gevisualiseerd door f kanluorescence microscopie met behulp van live of vaste cellen 6,7,8. Vergeleken met andere PCA&#39;s, BiFC is gevoelig en minder technisch uitdagend, met potentieel om de cellulaire lokalisatie in levende cellen te bestuderen. Een belangrijk nadeel van deze techniek is echter dat eenmaal gevormd, het fluorescerende eiwit complex kan niet worden omgekeerd. Daarom is het niet een goede methode om dynamische eiwit-eiwit interacties te bestuderen. In dit artikel gebruiken we BiFC om de interactie plaatsen van PDE4D3 kaart met AKAP-LBC door monitoring van de fluorescentie-intensiteiten van Venus. Vergeleken met traditionele analyse met fluorescentie microscopie, hetgeen tijdrovend en arbeidsintensief (indien niet uitgevoerd met behulp van geautomatiseerde high throughput machines), flowcytometrie biedt een eenvoudige kwantitatieve analyse van duizenden cellen in een heterogene populatie over een korte tijd 9, 10. Hier, door het uitvoeren van een side-by-side vergelijking van flowcytometrische analyse-BiFC en traditionele co-IP, tonen we aan dat de twee methoden bieden vergelijkbare gegevens echter flowcytometrische BiFC analyse is minder tijdrovend en gebruikt minder materiaal dat nuttiger kunnen zijn wanneer de cellen van belang zijn beperkt.

flow cytometric analysis of bimolecular fluorescence complementation: a high throughput quantitative method to study protein-protein interaction

Onder methodes om eiwit-eiwit interacties binnen cellen te bestuderen, Bimoleculaire Fluorescentie Complementatie (BiFC) is relatief eenvoudig en gevoelig. BiFC is gebaseerd op de productie van fluorescentie met behulp van twee niet fluorescerende fragmenten van een fluorescent eiwit (Venus, een geel fluorescent eiwit variant wordt hier gebruikt). Niet-fluorescerende Venus fragmenten (VN en VC) gefuseerd aan twee interagerende proteïnen (in casu AKAP LBC-en PDE4D3), waardoor fluorescentie door VN-AKAP-LBC-VC-PDE4D3 interactie en de vorming van een functioneel fluorescent eiwit in cellen. BiFC geeft informatie over de subcellulaire lokalisatie van eiwitcomplexen en de kracht van eiwit interacties op basis van fluorescentie-intensiteit. Echter, BiFC microscopische analyse van de sterkte van eiwit-eiwit interacties te kwantificeren is tijdrovend en enigszins subjectief door heterogeniteit in eiwitexpressie en interactie. Door koppeling flowcytometrischeanalyse BiFC methodologie, kan het fluorescerende BiFC eiwit-eiwitinteractie signaal nauwkeurig worden bepaald voor een grote hoeveelheid cellen in een korte tijd. Hier tonen we een toepassing van deze methode in kaart regio&#39;s in PDE4D3 die nodig zijn voor de interactie met AKAP-LBC. Deze high throughput methode kan worden toegepast op screening factoren die eiwit-eiwit interactie te bepalen.

AKAP-LBC en PDE4D3 constructies (Figuur 1B) werden gefuseerd met VN en VC fragmenten, respectievelijk. Interactie van AKAP-LBC en PDE4D3 resulteert in functionele YFP fluorescentie (Figuur 1A). Hier gebruiken we de BiFC methode om de kaart AKAP-LBC-bindingsplaatsen in PDE4D3.
