Kwantitatieve karakterisering van affiniteit van intermoleculaire interacties is belangrijk in veel gebieden van het biomedisch onderzoek. Binding dissociatieconstante (KD) is niet alleen noodzakelijk drug discovery, maar is ook een belangrijke parameter in de karakterisering van een binaire interactie in een biologisch systeem. Biochemische methoden voor detectie van eiwit-eiwit interacties, zoals immunoprecipitatie en gist twee-hybride-screening, niet aan ons van hoe strak zijn die interacties, terwijl affiniteit bepaald of dit complex bestaat onder bepaalde omstandigheden in vivo. In drug discovery proces, bindingassay ontwikkeling is een van de noodzakelijke en vaak het meest tijdrovende stappen. Meest gebruikte methoden van Kd bepaling omvatten fluorescentiepolarisatie, 1 surface plasmon resonance (SPR) technologie, 2 radioligand binding, 3 isothermische titratie calorimetrie 4 Equilibrium dialyse (ED), 5 ultrafiltratie (UF), 5 en ultracentrifuge (UC). 6 Allemaal vereisen aanzienlijke hoeveelheden gezuiverd doelwit eiwit. Microschaal thermophoresis (MST) is een zich snel ontwikkelende methode die gerichte beweging van moleculen in een microscopische temperatuurgradiënt detecteert. Eventuele wijzigingen van de hydratatie schil van biomoleculen resultaat in een relatieve verandering van beweging langs de temperatuurgradiënt. 7 MST wordt gebruikt om bindende affiniteiten te bepalen en is aangevraagd voor het bestuderen van ligand binding aan fluorescent gelabelde eiwitten of fluorescerende liganden aan een doelwit eiwit. 8, 9 MST maakt meting van interacties direct in oplossing zonder immobilisatie op een oppervlak (immobilisatie-vrije technologie). Praktisch wordt bindende gepaard met een verandering in het signaal MST, hoewel de grootte van de verandering verschilt van systeem tot systeem sterk. Voor de detectie molecuul verzoek van MST, zij moet fluorescerennt. Deze belangrijke beperking van de methode kan worden omgezet in een voordeel. Wanneer een eiwit tot expressie als een GFP-fusie in een systeem, zal de enige fluorescent molecuul zijn en dus kunnen worden bestudeerd zonder isolatie van het cellysaat of celvrij expressiesysteem. Generatie van cellysaten die het mogelijk maken voor bindende voorwaarden met minimale artefacten is de grote uitdaging. Hier een protocol cellysaat preparaat en MST experiment dat kan worden gebruikt voor vele oplosbare en membraaneiwitten beschrijven we. STAT eiwitten latente cytoplasmische transcriptiefactor geactiveerd door tyrosine fosforylering in respons op extracellulaire signalen en zijn betrokken bij vele biologische processen zoals immuniteit, hematopoiese, ontsteking en ontwikkeling. 10 In zoogdieren, de STAT STAT familie bestaat uit 1, 2, 3, 4 , 5A, 5B en 6. Alle geactiveerde STAT&#39;s zijn bekend te binden aan dezelfde DNA-sequentie, zogenaamde GAS motief, IFN-gamma geactiveerd sequentie. De transcriptioneel effecten van verschillende STAT&#39;s zijn zeer verschillend. 11 Ondanks betrokkenheid bij vele pathologische processen en uitgebreide studies waarbij meer dan 17.000 publicaties hebben de KD van STAT interacties met andere DNA-sequenties niet vastgesteld. Slechts relatieve affiniteit van verschillende statistieken tot varianten van GAS motief werd gekenmerkt. 11 Moeilijkheden in eiwit expressie en zuivering zijn de belangrijkste belemmeringen in de karakterisering van STAT &#39;DNA-binding selectiviteit. Hoewel de meeste studies zijn gericht op de rol van &quot;geactiveerde&quot; STAT, die synoniem geworden aan Tyr-gefosforyleerde transcriptiefactor, de rol van niet-gefosforyleerde STAT (U-STAT) in regulatie van transcriptie snel opkomende. 12 echter deze mechanismen worden slecht begrepen, en het was niet duidelijk of U-STAT daadwerkelijk binden aan DNA of optreden via interacties met andere transcriptiefactoren. We hebben onlangs aangetoond dat de U-STAT3 kan binden aan DNA sequences verschillend van GAS motieven met een nog hogere affiniteit. 13 De bevinding heeft belangrijke implicaties voor ons begrip van de biologische functies van deze belangrijke proteïne. We hebben microschaal thermophoresis relatieve affiniteiten van STAT3 bepalen GAS toegepast en AT-rijke oligonucleotide S 100 (figuur 4). Bijna identieke protocol is gebruikt KD bepaling voor binding van een ander ligand STAT3, een lipopeptide inhibitor. Nr. 14 binding aan een verbonden transcriptiefactor GFP-STAT1 die werd gebruikt als een negatieve controle worden gedetecteerd hetgeen bevestigt selectiviteit van interactie. 14

Kwantitatieve karakterisering van eiwit interacties is essentieel in vrijwel elk gebied van life sciences, met name drug discovery. De meeste momenteel beschikbare methoden Kd bepaling is toegang tot gezuiverde eiwit van belang, generatie die kan tijdrovend en duur. We hebben een protocol dat het mogelijk maakt voor de bepaling van de bindingsaffiniteit door microschaal thermophoresis (MST) zonder zuivering van het doelwit eiwit uit cellysaten ontwikkeld. De werkwijze omvat overexpressie van GFP gefuseerde eiwit en cellysis in niet-denaturerende omstandigheden. Toepassing van de methode STAT3-GFP transiënt tot expressie in HEK293 cellen zelf te bepalen voor de eerste maal de affiniteit van de goed onderzochte transcriptiefactor aan oligonucleotiden met verschillende sequenties. Het protocol is eenvoudig en kan een verscheidenheid aan applicatie voor het bestuderen van interacties van eiwitten met kleine moleculen, peptiden, DNA, RNA, en proteins.

Het meten van de affiniteit van de niet-gefosforyleerde STAT3 eiwit binding aan oligonucleotiden. HEK293 cellen die STAT3-GFP werden gebruikt als een bron van fluorescent gelabelde STAT3 voor DNA-bindingstest. Cellysaten werden bereid met RIPA buffer (20x10 6 cellen / ml).
