Prostaatkanker (PRCA) en benigne prostaathyperplasie (BPH) zijn significant gezondheidslasten. PRCA is de tweede meest voorkomende solide orgaantransplantatie kanker bij mannen en een belangrijke oorzaak van kanker-gerelateerde sterfte 1. BPH is ook zeer vaak voor bij oudere mannen, en er wordt geschat dat de klinische manifestatie van BPH, de lagere urinewegen (LUTS), is van invloed op 50-90% van de mannen 2. Terwijl androgenen en oestrogenen zijn bekend belangrijk om in PRCA en BPH, ons begrip van de hormonale mechanismen die ten grondslag liggen aan het ontstaan ​​van kanker en de groei is nog onvolledig 3,4. Eenvoudige en genetisch handelbaar diermodellen ondersteunen de studies die deze prioriteiten zullen met prostaat onderzoek. In hormoon reagerende aandoeningen zoals PRCA en BPH, het gebruik van subcutane, langzame afgifte hormoon pellets, op zich of in combinatie met een renale capsule xenografting (de overdracht van cellen, weefsels of organen van de ene soort in de andere) in het immunocompromised muisgastheer, biedt een eenvoudige en reproduceerbare methode voor het bestuderen van hormonale regulatie van de groei en carcinogenese. Subcutaan hormoon pellet implantatie is een eenvoudige en reproduceerbare techniek om hormonale regulatie van carcinogenese en goedaardige groei van de prostaat 5 bestuderen. renal capsule xenografting and subcutaneous pellet implantation for the evaluation of prostate carcinogenesis and benign prostatic hyperplasia

Nieuwe therapieën voor twee veel voorkomende prostaataandoeningen, prostaatkanker (PRCA) en benigne prostaathyperplasie (BPH), afhankelijk zijn van de experimenten evalueren van hun hormoonhuishouding. Sex steroïde hormonen (met name oestrogenen en androgenen) zijn belangrijk in PRCA en BPH, we onderzoeken hun respectieve rol in het veroorzaken van de groei prostaat en carcinogenese bij muizen met experimenten met gecomprimeerde hormoon pellets. Hormonen en / of geneesmiddel pellets gemakkelijk vervaardigd met een korrelpers, chirurgisch geïmplanteerd in het subcutane weefsel van de mannelijke muizengastheer. We een protocol beschrijven ook voor de evaluatie van hormonale carcinogenese door het combineren van subcutane hormoon pellet implantatie met xenografting van prostaat cel recombinanten onder de renale capsule van immuungecompromitteerde muizen. Bovendien subcutane hormoon pellet implantatie in combinatie met renale capsule xenografting van BPH weefsel, nuttig voor een beter begrip hormonale regulatie van goedaardige prostaten groei, en nieuwe behandelingen die sex steroïde hormoon pathways testen.
