Le tissu adipeux pour l&#39;isolement des CSA peut prendre de nombreuses formes: à partir de morceaux de tissus solides obtenus par résection ou lipoplasty de petits morceaux obtenus soit par extraction de la seringue ou lipoplastie assistée par aspiration (c&#39;est à dire la liposuccion). Que plus de cellules SVF (et donc CSA) peuvent être obtenus à partir d&#39;échantillons de tissu adipeux résection ou aspiration n&#39;est pas clair que les études contradictoires ont été présentées 16, 17....

En 2001, une population présumée de cellules souches multipotentes à partir de tissu adipeux a été décrite dans la revue Tissue Engineering 1. Ces cellules ont été donné le nom de cellules transformés lipoaspirat ou PLA en raison de leur dérivation à partir de tissu de lipoaspirat traités obtenus par la chirurgie esthétique. Le procédé d&#39;isolement décrit dans cet article est basée sur des stratégies existantes enzymatiques pour l&#39;isolement de la fraction stroma vasculaire (SVF) à partir de tissu adipeux 2. Le SVF a été définie comme une transformation minimale population de globules rouges, les fibroblastes, les cellules endothéliales, les cellules musculaires lisses, des pericytes et des pré-adipocytes qui restent encore à adhérer à un substrat de culture de tissus 2, 3. La culture de cette SVF dans le temps est proposé d&#39;éliminer bon nombre de ces populations de cellules contaminantes et entraîner, une population de fibroblastes adhérent. Ces fibroblastes ont été identifiés dans la littérature pour les 40 dernières années comme étant pré-adipocytes. Toutefois, notre groupe de recherche a démontré que ces cellules possédaient mésodermique multipotency et renommés de la population SVF adhérente que les cellules de l&#39;APL. Des études ultérieures par de nombreux autres groupes de recherche ont ajouté à ce potentiel, suggérant à la fois endodermique et potentiels ectodermiques (pour revue, voir 4). Depuis ce temps, de nombreux termes supplémentaires pour ces cellules ont été publiés dans la littérature. Afin de fournir un certain type de consensus, le terme de cellules souches du tissu adipeux ou CSA a été adopté lors de la deuxième conférence annuelle IFATS 2. En tant que tel, l&#39;ASC terme sera utilisé dans cet article. Le protocole décrit dans cet article est une procédure relativement simple qui nécessite un équipement de laboratoire standard et utilise des réactifs simples, telles que la phospho-solution saline tamponnée, des réactifs de milieux de culture de tissus standard et de la collagénase. Il peut produire un grand nombre de CSA en fonction de la quantité de départ de volume du tissu adipeux et c ultérieurulture temps. Cependant, le traitement d&#39;une telle quantité de tissu adipeux peut présenter certains problèmes physiques qui peuvent être atténués à un degré en utilisant ce protocole. En outre, ce protocole ne nécessite tissus stériles installations de culture et les hottes de biosécurité approuvées, ce qui nécessite l&#39;utilisation d&#39;une installation de culture de tissus approuvé. Cette exigence peut également diminuer l&#39;utilité de la population de l&#39;ASC dans les applications cliniques, sauf s&#39;ils sont isolés dans de bonnes pratiques de fabrication des installations (GMP) approuvés conçus pour l&#39;isolement et l&#39;expansion des matériaux pour une utilisation clinique. Comme alternative, les systèmes automatisés qui peuvent isoler CSA dans un système fermé dans la salle d&#39;opération permettrait d&#39;éviter cette question clé et permettre l&#39;utilisation immédiate des CSA sans qu&#39;il soit nécessaire pour l&#39;expansion in vitro ultérieure. A ce jour, il existe six systèmes automatisés qui sont commercialement disponibles pour l&#39;isolement de cellules à partir de tissu humain. Ces systèmes peuvent permettre d&#39;isoler unnombre important des CSA de grandes quantités de tissu adipeux immédiatement après sa récolte. Ces CSA pourraient alors être réintroduits dans le patient pour une variété de fins de régénération sans que le patient jamais avoir à quitter la salle d&#39;opération. En plus de ce protocole décrivant l&#39;isolement d&#39;emploi des CSA, un protocole pour l&#39;isolement automatique des CSA à l&#39;aide du système Celution est également donnée dans un article associé.

En 2001, des chercheurs de l&#39;Université de Californie, Los Angeles, décrit l&#39;isolement d&#39;une nouvelle population de cellules souches adultes à partir de tissu adipeux que Liposuctioned qu&#39;ils appellent d&#39;abord les cellules de lipoaspirat transformés ou des cellules de l&#39;APL. Depuis lors, ces cellules souches ont été renommés que les cellules souches dérivées de tissu adipeux ou CSA et ont continué pour devenir l&#39;un des plus populaires des populations les cellules souches adultes dans les domaines de la recherche sur les cellules souches et la médecine régénérative. Des milliers d&#39;articles décrivent maintenant l&#39;utilisation des CSA dans une variété de modèles animaux de régénération, y compris la régénération osseuse, réparation des nerfs périphériques et de l&#39;ingénierie cardiovasculaire. Des articles récents ont commencé à décrire la multitude d&#39;utilisations pour CSA dans la clinique. Le protocole indiqué dans cet article décrit la procédure de base pour la main et enzymatique isoler CSA de grandes quantités de lipoaspirates obtenus à partir des procédures cosmétiques. Ce protocole peut être facilement agrandie ou réduite à accommodmangé le volume du lipoaspirat et peut être adapté pour isoler à partir de tissu adipeux ASCs obtenus par plasties abdominales et d&#39;autres procédures similaires.

Le contour du protocole décrit ci-dessus, un procédé enzymatique manuel pour l&#39;isolement d&#39;un SVF à partir d&#39;un grand échantillon de lipoaspirat de volume. Dans ce SVF sont nombreuses populations de cellules, y compris l&#39;ASC.
