Em 2001, pesquisadores da Universidade da Califórnia, em Los Angeles, descreveu o isolamento de uma nova população de células-tronco adultas do tecido adiposo lipoaspirada que inicialmente denominado Células lipoaspirado ou células processados ​​PLA. Desde então, essas células-tronco foram renomeados como células-tronco adiposo-derivadas ou ASC e passaram a se tornar um dos mais populares populações de células-tronco adultas nos campos da investigação em células estaminais e medicina regenerativa. Milhares de artigos agora descrever o uso de ASC em uma variedade de modelos animais regenerativos, incluindo a regeneração óssea, reparação de nervos periféricos e engenharia cardiovascular. Artigos recentes começaram a descrever a miríade de usos para ASC na clínica. O protocolo mostrado neste artigo descreve o procedimento básico para manualmente e enzimaticamente isolando ASC de grandes quantidades de lipoaspirates obtidos a partir de procedimentos cosméticos. Este protocolo pode ser facilmente escalado para cima ou para baixo para accommodcomeu o volume de lipoaspirado e pode ser adaptado para isolar as ASC de tecido adiposo obtidas por abdominoplasties e outros processos semelhantes.

O esboço protocolo acima descreve um método manual, enzimática para o isolamento de um SVF a partir de um grande volume de amostra lipoaspirado. Dentro deste SVF são populações de células numerosas, incluindo o ASC. Numerosos estudos propõem que este SVF cultura sob condições de cultura de tecidos padrão irá seleccionar uma população de fibroblastos aderentes susceptíveis de ser composto principalmente do tipo ASC.
