Fettvävnad för isolering av ASCs kan komma i många former: från fasta bitar av vävnad som framställts genom resektion eller lipoplasty till mindre bitar som erhållits genom antingen spruta extraktion eller sug-assisterad lipoplasty (dvs. fettsugning). Huruvida fler SVF celler (och därmed ASCs) kan erhållas från opererande eller aspirefettprover är oklart motstridiga studier har presente 16, 17. År 2001 var en förmodad befolkning på multipotenta stamceller från fettvävnad som beskrivs i tidskriften Tissue Engineering 1. Dessa celler fick namnet Bearbetade Lipoaspirate eller PLA-celler på grund av deras härledning från bearbetad lipoaspirate vävnad som erhållits genom plastikkirurgi. Isoleringen metod som beskrivs i den här artikeln byggde på befintliga enzymatiska strategier för isolering av stromaceller vaskulära fraktion (SVF) från fettvävnad 2. Den SVF har definierats som ett minimalt bearbetade population av röda blodceller, fibroblaster, endotelceller, glatta muskelceller, pericyter och pre-adipocyter som ännu inte hålla sig till en vävnadsodlingssubstrat 2, 3. Odling av denna SVF över tiden har föreslagits för att eliminera många av dessa kontaminerande cellpopulationer och resulterar i en vidhäftande, fibroblastisk befolkning. Dessa fibroblaster har identifierats i litteraturen för de senaste 40 åren som är pre-adipocyter. Men vår forskargrupp visat att dessa celler hade mesodermal multipotency och döptes den vidhäftande SVF befolkningen som PLA-celler. Efterföljande studier av ett stort antal andra forskargrupper har lagt till denna potential, vilket tyder både endodermalt och ektodermala potentialer (för översikt se 4). Protokollet beskrivs i denna artikel är en relativt enkel procedur som kräver standard laboratorieutrustning och använder enkla reagenser såsom fosfor-saltlösning, standardvävnadsodlingsmedier reagenser och kollagenas. Den kan producera ett stort antal DSKer beroende på mängden av utgångs fettvävnad volym och efterföljande cKULTUR tid. Emellertid kan behandling av en sådan stor mängd av fettvävnad presentera några fysiska problem som kan minskas till en grad med hjälp av detta protokoll. Dessutom går detta protokoll kräver sterilt odlingsanläggningar och godkända biosäkerhet huvor, vilket nödvändiggör användning av en godkänd vävnadskultur anläggning. Detta krav kan också minska nyttan av ASC populationen i kliniska tillämpningar, om de är isolerade i god tillverkningssed (GMP)-godkända anläggningar avsedda för isolering och expansion av material för klinisk användning. Som ett alternativ, skulle automatiserade system som kan isolera ASCs i ett slutet system i operationssalen undvika denna viktiga fråga och möjliggöra omedelbar användning av ASCs utan behov av efterföljande in vitro expansion. Till dags dato finns det sex automatiserade system som är kommersiellt tillgängliga för isolering av celler från human vävnad. Dessa system kan göra det möjligt att isolera enbetydande antal ASCs från stora mängder fettvävnad omedelbart efter sin skörd. Dessa ASCs kan sedan återinföras i patienten för olika regenerativa ändamål utan patienten någonsin behöva lämna operationssalen. manual isolation of adipose-derived stem cells from human lipoaspirates

År 2001, forskare vid University of California, Los Angeles, beskrev isoleringen av en ny population av adulta stamceller från fettsugning fettvävnad att de initialt kallas Bearbetade Lipoaspirate Celler eller PLA-celler. Sedan dess har dessa stamceller döpts som Fett-härledda stamceller eller ASCs och har gått vidare till att bli en av de mest populära vuxna stamceller populationer inom områdena stamcellsforskning och regenerativ medicin. Tusentals artiklar beskrivs nu användningen av ASC: er i en mängd olika regenerativa djurmodeller, inklusive benregenerering, perifer nervreparation och hjärt-engineering. Nya artiklar har börjat för att beskriva den mängd olika användningsområden för ASCs i kliniken. Det protokoll som visas i den här artikeln beskriver den grundläggande proceduren för manuell och enzymatiskt isolera ASCs från stora mängder lipoaspirates erhållits från kosmetiska behandlingar. Detta protokoll kan enkelt skalas upp eller ner för att accommodåt volymen lipoaspirate och kan anpassas för att isolera DSKer från fettvävnad erhållna genom abdominoplasties och liknande förfaranden.
