Microfluidics technologie heeft nieuwe en fascinerende mogelijkheden voor single-cell analyse. Vele studies in het verleden cel tot cel verschillen binnen een grote celpopulatie 1-3, in het bijzonder signalering verwerkt 4 of de hoeveelheden intracellulaire biomoleculen zoals eiwitten 5,6, metabolieten en cofactoren 7,8 geopenbaard. Deze heterogeniteit wordt geacht fundamenteel belang voor de aanpassing en evolutie 9 cellen toe, maar spelen ook een belangrijke rol bij het ​​ontstaan ​​en behandeling van ziekten zoals kanker 10-13. Daarom studies over de eencellige niveau van groot belang in biologisch en farmacologisch onderzoek, vooral als deze studies tonen de verschillende celreacties na behandeling met bioactieve chemicaliën. De laatste jaren zijn veel analytische platforms ontwikkeld die de analyse van afzonderlijke levende cellen of de chemische samenstelling van de celinhoud vergemakkelijken. Terwijl fluorescent geactiveerde cel sortering (FACS) is de gouden stAndard voor hoge-doorvoer analyse van afzonderlijke levende cellen, de methode niet worden toegepast voor de kwantificering van intracellulaire of uitgescheiden verbindingen. De opkomst van microfluïdische platforms heeft nieuwe analytische strategieën voor de positionering, de behandeling en observatie van enkele cellen beloofd. Een mijlpaal in microfluidics werd bereikt met de integratie van flexibele PDMS kleppen gerealiseerd door Quake en collega&#39;s 14,15. Deze kleppen zijn nuttig omdat ze regio&#39;s kunnen isoleren op chip, bijv. scheiden twee culturen 16. De kracht van deze aanpak voor single-cell analyse is onlangs aangetoond door Hansen en collega&#39;s, die de genexpressie geanalyseerd uit honderden individuele cellen in parallel 17. Bij targeting eiwitten en metabolieten, de analyse moeilijk door het ontbreken van een geschiktemplification methoden, het grote aantal verschillende verbindingen aanwezig zijn, en de verschillen in chemische aard. Bovendien zijn de meeste intracellulaire biomoleculen verwacht aanwezig in lage kopie-aantallen in de orde van enkele tienduizenden 18 te zijn, vandaar de gebruikte analysemethode moet een hoge gevoeligheid hebben. Krachtiger testen zoals immunoassays en enzym-linked immunoassays (ELISA) zijn moeilijk te integreren in microfluïdische apparaten omdat ze vereisen een aantal wassen en incubatie stappen evenals oppervlak immobilisatie. Als gevolg van deze uitdagingen, is het niet verwonderlijk dat slechts een paar voorbeelden zijn gemeld waar eiwitten of metabolieten werden gekwantificeerd op de single-cell niveau. Zo hebben studies naar de uitscheiding van fluorescerende verbindingen gemeld 19,20. Onlangs heeft de uitvoering met ELISA werd voor de analyse van uitgescheiden (niet-fluorescerende) eiwitten van een celkweek (THP-1-cellen) 21 en single (immune) cellen 10. Targeting intracellulaire eiwitten, Shi et al.. Ontwikkelde een microfluïdische apparaat dat de identificatie van intracellulaire eiwitten voor de analyse van signaal transductie in tumorcellen door middel van een immunoassay 11 vergemakkelijkt. Slechts relatieve hoeveelheden eiwitten werden bepaald en geen enzymatische amplificatie werd gebruikt om het signaal voor lage abundantie proteïnen verhogen. Onlangs waren we in staat om een single-cell trapping microdevice met fluorescentie-assays 8 en immunoassays 22 (figuur 1) te combineren. Cellen worden passief gevangen in microsized hindernis structuren, die het aanbod en (snelle) uitwisseling van middelgrote en andere chemische middelen zonder enige beweging van de cellen mogelijk te maken. Een ringvormige klep rond elke val maakt isolatie van de cel in een zeer klein volume (&quot;de microkamer&quot;). Deze klep wordt geactiveerd direct na de invoering van een cel-lyseren (hypoosmolar) buffer, hijNVU voorkomen intracellulaire moleculen of uitgescheiden moleculen weg te diffunderen. Het belangrijkste is, vanwege de kleine grootte van het volume (625 pl) grote verdunning van de moleculen wordt vermeden. Aangezien lysis en analyse uitgevoerd op dezelfde positie in de chip, is er geen verlies van analyten door transport. Tot nu toe hebben we de inrichting voor de analyse van intracellular en uitgescheiden enzymen (relatieve kwantificatie door enzymatische assays), intracellulaire cofactoren, eiwitten en kleine moleculen (absolute kwantificatie door eindpunt assays of ELISA). Hieronder beschrijven we de werkwijze chip fabricage door middel van zachte meerlaagslithografie en protocollen voor patroonvorming van de antilichamen via microcontact printen en oppervlaktechemie. Daarnaast is een aantal voorbeelden van de chip gebruik en behandelingen worden gegeven.

a microfluidic chip for the versatile chemical analysis of single cells

Wij presenteren een microfluïdische apparaat dat de kwantitatieve bepaling van intracellulaire biomoleculen in meerdere afzonderlijke cellen parallel maakt. Hiertoe worden de cellen passief gevangen in het midden van een microkamer. Na activering van de besturingslaag wordt de cel geïsoleerd van de omringende volume in een kleine kamer. De omliggende volume kan dan worden uitgewisseld zonder dat de geïsoleerde cel. Echter, bij korte openen en sluiten van de kamer, de oplossing in de kamer kan worden vervangen binnen een paar honderd milliseconden. Door de omkeerbaarheid van de kamers kunnen de cellen worden blootgesteld aan verschillende oplossingen achtereenvolgens een zeer controleerbare wijze, bijv. voor incubatie, wassen en tenslotte cellysis. De goed afgesloten microchambers staat het behoud van het lysaat, minimaliseren en de controle van de verdunning na cellysis. Aangezien lysis en analyse plaatsvinden op dezelfde locatie, is hoge gevoeligheid behouden omdat geen verdere dilution of verlies van de analyten optreedt tijdens het transport. De microkamer ontwerp maakt daarom de betrouwbare en reproduceerbare analyse van zeer kleine kopie-aantallen van intracellulaire moleculen (attomol, zeptomol) vrijgelaten uit individuele cellen. Bovendien kunnen vele microchambers worden opgesteld in een matrix format, waarbij de analyse van vele cellen tegelijk, aangezien geschikte optische instrumenten worden gebruikt voor bewaking. We hebben al gebruik van het platform voor proof-of-concept studies naar intracellulaire eiwitten, enzymen, co-factoren en second messengers in zowel relatieve of absolute meetbare wijze te analyseren.

Het platform kan een aantal intracellulaire en afgescheiden moleculen in of geproduceerd door enkele cellen te analyseren. Hier willen we ander voorbeeld studies aan de verscheidenheid van mogelijke tests onderstrepen. We zullen een voorbeeld voor een uitgescheiden enzym (figuur 5a) en een intracellulair enzym (figuur 5b) en eiwit (Figuren 5c en d).
