Zoogdieren spermatogenese is een complex proces dat differentiatie in verschillende stadia van de testes van de testes 1. Gelukkig is de spermatogenese celtypen sterk verschillen in omvang, waardoor voor een enkele-celsuspensie verkregen uit geheel testes met een gradiënt vloeistof te scheiden. De STA-PUT methode, hier aangetoond, maakt gebruik van een lineaire gradiënt BSA en eenvoudig sedimentatie te spermatogene cellen te scheiden op basis van grootte en massa 6-9. De STA-PUT methode heeft een aantal voordelen ten opzichte van de andere twee meest gebruikte methoden om spermatogene celtypes scheiden: FACS en elutriatie 10-13. De STA-PUT apparaat vereist only verschillende stukken van gespecialiseerde glaswerk geassembleerd in een koude kamer of grote koelkast. Zo is goedkoper dan het gebruik van een cel sorteerder of een afslibinrichting. De STA-PUT methode levert grotere hoeveelheden cellen per celtype en testis dan kan worden gesorteerd door FACS op een vergelijkbare tijdsspanne, hoewel de zuiverheid van elke celpopulatie is niet zo hoog als die verkregen met FACS 11. Celsortering gebruik magnetische korrels (magnetisch geactiveerde celsortering, MACS) is recent ook succesvol toegepast voor verrijking spermatogonia uit een gemengde testicular celpopulatie, maar het is nog niet geschikt voor het scheiden spermatocyten of spermatiden door gebrek aan kennis van geschikte oppervlakte-markers 14. Een bijkomend voordeel van de STA-PUT methode via FACS of MACS is de mogelijkheid om levende cellen voor verdere kweek isoleren omdat in tegenstelling tot de meeste FACS protocollen, het geen DNA of andere vormen van vlekken vereisen. Voor studies die ik nodigarge opbrengsten van spermatogene cellen vormen op ~ 90% zuiverheid, de STA-PUT is een ideale methode.

Zoogdieren spermatogenese is een complex differentiatie proces dat plaatsvindt in verschillende stadia in de testes van de testes. Momenteel is er geen betrouwbare celkweeksysteem waardoor spermatogene differentiatie in vitro, en de meeste biologische studies van spermatogene cellen vereisen weefsel oogst uit dierlijke modellen zoals de muis en rat.
