La méthode STA-PUT, démontré ici, utilise un gradient linéaire BSA et simple sédimentation pour séparer les cellules spermatiques fonction de la taille et de la masse 6-9. La méthode STA-PUT produit des quantités plus élevées de cellules par type de cellule et les testicules que peuvent être triés par FACS dans un laps de temps comparable, bien que la pureté de chaque population de cellules n&#39;est pas aussi élevés que ceux obtenus avec FACS 11. Tri cellulaire utilisant des billes magnétiques (magnétique tri cellulaire activé, MACS) a récemment été utilisée avec succès pour l&#39;enrichissement de spermatogonies à partir d&#39;une population de cellules testiculaires mixte, mais il est actuellement inadapté pour séparer spermatocytes ou spermatides en raison du manque de connaissances de surface appropriée les marqueurs 14. Un avantage supplémentaire de la méthode STA-PUT sur FACS ou MACS est la capacité à isoler des cellules viables aptes à la culture ultérieure parce que, contrairement à la plupart des protocoles de FACS, il ne nécessite pas d&#39;ADN ou d&#39;autres types de coloration. Spermatogenèse chez les mammifères est un processus de différenciation complexe qui se produit en plusieurs étapes dans les tubules séminifères des testicules. Actuellement, il n&#39;existe pas de système de culture cellulaire fiable permettant la différenciation de la spermatogenèse in vitro, et la plupart des études biologiques des cellules spermatiques nécessite récolte des tissus de modèles animaux comme la souris et le rat.
