Däggdjurs spermatogenesen är en komplex differentiering process som sker i flera steg i sädeskanalerna i testiklarna 1. Kortfattat, stem-like spermatogonier att uppe nära epitelet sädeskanalens tubulus klyftan och differentiera till spermatocyter, vilka sedan genomgår meiotiska delningar. Efter meios är klar, de resulterande haploida celler, eller runda spermatider, genomgår spermiogenesen, en differentieringsprocess som involverar avgivande av cytoplasman och kompaktering av kärnan. Spermatider successivt utveckla en flagellum och genomgå töjning och kondensering av kärnan, som producerar förlängning och sedan kondensespermatider, respektive. Cellsortering utnyttjar magnetiska kulor (magnetisk aktiverad cellsortering, MACS) har nyligen framgångsrikt använts för anrikning av spermatogonier från en blandad testikelcellpopulation, men det är för tillfället olämpligt för att separera spermatocyter eller spermatider på grund av bristande kunskap om lämplig yta markörer 14. En ytterligare fördel med STA-PUT-metod under FACS eller MACS är förmågan att isolera livsdugliga celler som är lämpliga för efterföljande kultur, eftersom det i motsats till de flesta FACS-protokoll, behöver den inte någon DNA-eller andra typer av färgning. Däggdjurs spermatogenesen är en komplex differentiering process som sker i flera steg i sädeskanalerna i testiklarna. För närvarande finns det inga tillförlitliga cellodlingssystem möjliggör spermatogenesisk differentiering in vitro, och de flesta biologiska studier av spermatogenescykler celler kräver vävnad skörd från djurmodeller såsom mus och råtta. Även celler som isolerats från testiklarna är sedimente genom BSA lutning, kan observeras flera olika band av celler. Eventuella klumpar av celler som har en tendens att sjunka till botten av gradienten och kommer inte förorena de andra fraktionerna.
