Animals Used
This protocol has been optimized for Swiss Webster mice. However, this method is easily adaptable to other small-sized mammals such as rats and to other strains of mice. We have successfully performed preliminary isolations with C57 mice and &mu;-opioid receptor knock-outs. Das enterische Nervensystem (ENS) ist groß Netzwerk von Nerven und Gliazellen, die die gesamte Länge des Magen-Darm-Trakt (GI) verläuft. an in-vitro preparation of isolated enteric neurons and glia from the myenteric plexus of the adult mouse

Die enterale Nervensystem ist ein riesiges Netz von Neuronen und Gliazellen, die die Länge des Magen-Darm-Trakt, die funktionell steuert gastrointestinalen Motilität. Ein Verfahren zur Isolierung und Kultivierung von einer gemischten Population von Neuronen und Gliazellen aus dem Plexus myentericus beschrieben. Die primären Kulturen kann für mehr als 7 Tage gehalten werden, mit Verbindungen zwischen den Neuronen entwickeln und Gliazellen. Die Längsmuskel Streifen mit dem beigefügten myentericus Plexus aus dem zugrunde liegenden kreisförmigen Muskel der Maus Ileum oder Kolon abgestreift und einer enzymatischen Verdauung. In sterilen Bedingungen werden die isolierten neuronalen und Gliazellen Bevölkerung im Pellet nach Zentrifugation erhalten und plattiert auf Deckgläsern. Innerhalb von 24-48 Stunden, tritt Neuritenwachstum und Neuronen kann durch pan-neuronalen Marker identifiziert werden. Nach zwei Tagen in Kultur isolierten Neuronen feuern Aktionspotentiale durch Patch-Clamp-Studien beobachtet. Darüber hinaus können auch magensaftresistente Glia identif seinied von GFAP-Färbung. Ein Netzwerk von Neuronen und Gliazellen in enger Apposition bildet innerhalb von 5 - 7 Tage die. Enteric Neuronen können individuell und direkt untersucht mit Methoden wie Immunhistochemie, Elektrophysiologie, Kalzium-Imaging und Single-Cell-PCR. Darüber hinaus kann dieses Verfahren in genetisch veränderte Tiere durchgeführt werden. Diese Methode ist einfach durchzuführen und kostengünstig. Insgesamt macht dieses Protokoll die Komponenten des enteralen Nervensystems in einem leicht manipuliert Weise, so dass wir Ihnen besser entdecken Sie die Funktionalität der ENS in normalen und Krankheitszuständen.

Immediately following isolation of LMMP-derived cells, neurons and other cell types will not be readily evident. Living, round cells of indistinct phenotype can be seen as well as tissue detritus from incompletely digested tissue fragments and connective tissue.
