De bacteriën moet maken met herkenning en antimicrobiële activiteiten van het gastheerimmuunsysteem. De gastheer immuunsysteem bestaat uit aangeboren en adaptieve componenten, waaronder fysische en chemische barrières, immuuncellen, het complementsysteem, en andere onderdelen van humorale immuniteit. fluorescence microscopy methods for determining the viability of bacteria in association with mammalian cells

Centraal in het gebied van bacteriële pathogenese is de mogelijkheid om te bepalen of en hoe microben overleven na blootstelling aan eukaryotische cellen. Huidige protocollen om deze vragen te beantwoorden zijn onder kolonietelling assays, gentamicine protection assays en elektronenmicroscopie. Kiemgetal en bescherming gentamicine assays alleen de levensvatbaarheid van de totale bacteriële populatie te beoordelen en in staat zijn om individuele bacteriële levensvatbaarheid te bepalen. Elektronenmicroscopie kan worden gebruikt om de levensvatbaarheid van individuele bacteriën bepalen en informatie over hun lokalisatie in gastheercellen. Echter, bacteriën vertonen vaak een aantal elektron dichtheden, waardoor de beoordeling van de levensvatbaarheid moeilijk. Dit artikel beschrijft protocollen voor het gebruik van fluorescerende kleurstoffen die de levensvatbaarheid van individuele bacteriën in en gekoppeld aan gastheercellen onthullen. Deze testen werden oorspronkelijk ontwikkeld om de overleving van Neisseria gonorrhoeae beoordelen in primaire humane neutrofielen, maar moet eenpplicable aan een bacterie-gastheercel interactie. Deze protocollen gecombineerd membraan-permeabele fluorescente kleurstoffen (SYTO9 en 4 &#39;,6-diamidino-2-fenylindool [DAPI]), die alle bacteriën kleuren met membraan-ondoorlaatbare fluorescerende kleurstoffen (propidiumjodide en SYTOX Green), die alleen toegankelijk zijn levensvatbaar bacteriën. Vóór eukaryotische cel permeabilisatie, wordt een antilichaam of fluorescerend reagens extracellulaire bacteriën te identificeren. Dus deze assays discrimineren de levensvatbaarheid van bacteriën aanhanger en in eukaryotische cellen. Een protocol is ook voorzien voor gebruik van de levensvatbaarheid kleurstoffen in combinatie met fluorescente antilichamen tegen eukaryote cel markers om de subcellulaire lokalisatie van individuele bacteriën te bepalen. De bacteriële levensvatbaarheid kleurstoffen besproken in dit artikel zijn een gevoelig aanvulling en / of alternatief voor de traditionele microbiologie technieken om de levensvatbaarheid van de individuele bacteriën evalueren en informatie verstrekken over waar bacteriën overleven in gastheercels.

De geschetste protocollen werden gebruikt om overleving van N. onderzoeken gonorrhoeae na blootstelling aan primaire humane neutrofielen 5,26. Neutrofielen werden geïnfecteerd met N. gonorrhoeae en verwerkt met protocol 1, met behulp van de groene fluorescerende kleurstof SYTO9 levensvatbaarheid en de rood-fluorescerende propidiumiodide (Figuur 4A).
