MicroRNA zijn klein (ongeveer 22 nucleotiden lang) niet-coderende RNA&#39;s die endogeen wordt geproduceerd. Zij over het algemeen functioneren van eiwit expressie te onderdrukken door middel van translationeel repressie of mRNA degradatie. miRNAs binden aan mRNA doelen met onvolledige complementariteit, waardoor het mogelijk is voor een enkele miRNA aan verschillende doelen te onderdrukken. Begrijpen welke celtypen en structuren te uiten miRNA&#39;s is een belangrijk onderdeel van het begrijpen van de mechanismen waardoor veranderingen in miRNA expressie invloed cellen en weefsels fenotypes. Hoewel methoden zoals miRNA sequencing, qPCR en Northern blotting kan worden gebruikt voor de detectie van miRNAs geheel weefsels staat deze benadering niet mogelijk maken om te bepalen welke specifieke celtype ze uit in een gegeven weefsel. Dissectie van cellulaire en structurele componenten voorafgaand aan de analyse met behulp van deze methoden kan heel moeilijk zijn en het nodig is om een ​​adequate isolatie bereiken omstandigheden kunnen leiden tot alterations in genexpressie of afbraak van RNA&#39;s. microRNA in situ hybridisatie is een methode die wordt gebruikt om microRNA locatie en expressie niveaus visualiseren in weefsels. Deze techniek is bijzonder waardevol in weefsels bestaande uit heterogene structuren, zoals de nieren. MicroRNAs is aangetoond dat een regulerende rol in de ontwikkeling nier 2,3 en fysiologie 4 spelen. microRNA expressie veranderingen hebben ook aangetoond te worden betrokken met nier pathologie zoals fibrose 5-10, diabetische nefropathie 7, niercarcinoom 11,12, en acute nierschade 13. In ons onderzoek hebben wij gevonden dat het optimaliseren van microRNA in situ hybridisatie voor nierweefsel was waardevol voor het bepalen van de exacte locaties van structurele miRNA expressie in zowel gezondheid en ziekte 14. Bepaling van de buisvormige en cellulaire expressie van verschillende microRNAs is belangrijk omdat de regulering van targets kan afhankelijk cellulaire functies zijn. In zieke toestanden is het ook belangrijk om te bepalen hoe veranderingen in miRNA expressie kan beïnvloeden functie. Het doel van de hier beschreven methode was om voort te bouwen op bestaande ISH methodieken ontwikkeld door Kloosterman et al.. 15, andere onderzoekers 16,17, en die voorgesteld door Exiqon 1 en optimaliseren van de methode voor het formaline gefixeerde nierweefsel. We hebben met succes gebruik gemaakt van deze methode om duidelijke regionale verschillen in de renale microRNA miR-382 expressie met eenzijdige ureterobstructie 18 identificeren. In dit artikel wordt een methode voor de colorimetrische detectie van miRNA in de nier beschrijven we door in situ hybridisatie met digoxigenine gelabeld microRNA probes. Dit protocol, oorspronkelijk ontwikkeld door Kloosterman en collega&#39;s voor een breed gebruik met Exiqon miRNA sondes 1, is aangepast om de uitdagingen die inherent zijn miRNA analyse in nierweefsel overwinnen.
