I testilysatorer skiljer sig könsceller från diploid spermatogonia genom meios till mogna, haploid spermatozoa. Under denna process ombyggnads den nukleära kromatinstrukturen hos könsceller kontinuerligt och dynamiskt. Ytspridningar används ofta för cytologisk undersökning av enskilda spermatogena celler. En rådande metod för ytspridning använder hypoton behandling genom vilken enskilda spermatogena celler sprids och plattasut 1. Dessa villkor är optimala för detaljerad analys av meiotiska kromosomer. Kromosomala funktioner som synaptisk status och rekombinationsfoci observeras lätt med denna metod. Hypotona behandlingen stör dock subnukleära chromatin arkitektur, således denna teknik är inte lämplig för strukturell analys av atomkärnor. Följaktligen har en förbättrad metod utformats för att bevara den tredimensionella kromatinstrukturen hos testikelceller. Denna metod har kallats den tredimensionella (3D) bildmetoden (figur 1). 3D-glidmetoden har gjort det möjligt att upptäcka gryende RNA-lokalisering i atomkärnor eftersom den ursprungligen optimerades för att undersöka genuttryck och kromatin tillstånd under testikel könsceller differentiering av RNA fluorescens in situ hybridisering (FISH)2,3. Denna 3D-metod är också tillämplig på kombinationen av immunofluorescens, DNA och RNA FISH. Dessutom har denna metod hjälpt till att upptäcka postmeiotisk könskromatin (PMSC), ett tyst fack av könskromosomerna som finns i postmeiotiska spermatider2.
3D-glidmetoden optimerades ursprungligen genom kombinationen av två viktiga steg för glidberedning som vanligtvis används för kärnfärgning: fixeringssteget som utformats för att fixa kärnmaterial och permeabiliseringssteget avsett att ta bort cytoplasmaiska material för att förbättra tillgängligheten för färgreagenser, såsom antikroppar och FISK-sonder. Som tidigare beskrivits i enannan publikation 3har det fastställts att permeabiliseringssteget måste föregå fixeringssteget för att uppnå optimala resultat med låg cytoplasmisk bakgrund. I denna 3D-glidmetod utförs permeabiliseringssteget och efterföljande fixeringssteg direkt på seminiferous tubules och följs av mekanisk dissociation av könsceller med tång före cytospinning på diabilder. En alternativ metod för RNA FISH av spermatogenic celler utvecklades i ett annat laboratorium4. I denna metod, i överensstämmelse med 3D-metoden, måste permeabiliseringssteget föregå fixeringssteget för att uppnå optimala resultat av RNA FISH.
Följande protokoll beskriver 3D-bildmetoden och ger ett exempel på en möjlig tillämpning, Cot-1 RNA FISH för detektion av nukleära gryende RNAs. Cot-1 DNA består av repetitiva element i genomet. Här hybridiseras en Cot-1 DNA-sond till en intron och UTR av de gryende transkriptionerna, vilket detekterar de transkriptionellt aktiva regionerna i kärnan5,6. 3D-bilder är mångsidiga och kan appliceras på en kombination av immunofluorescens-, DNA- och RNA FISH-tekniker för att få detaljerad undersökning av rumsliga relationer mellan kromatinarkitektur.

Under testikel könsceller differentiering förändras strukturen av nukleära kromatin dynamiskt. Nedan beskrivs en metod som är utformad för att bevara det tredimensionella kromatinarrangemanget av testikelceller som finns hos möss. Denna metod har kallats den tredimensionella (3D) bildmetoden. I denna metod behandlas testikelrör direkt med ett permeabiliseringssteg som tar bort cytoplasmiskt material, följt av ett fixeringssteg som fixerar kärnmaterial. Tubuler demonteras sedan, cellupphängningen är cytospun och cellerna klibbar vid diabilder. Denna metod förbättrar känsligheten för detektion av subnukleära strukturer och är tillämplig för immunofluorescens, DNA och RNA fluorescens in situ hybridisering (FISH) och kombinationen av dessa detektionsmetoder. Som ett exempel på en möjlig tillämpning av 3D-glidmetoden visas en Cot-1 RNA FISH för att upptäcka gryende RNAs. 3D-glidmetoden kommer att underlätta detaljerad undersökning av rumsliga relationer mellan kromatinstruktur, DNA och RNA under testikelreceller differentiering.

I detta experiment utfördes Cot-1 RNA FISH för att upptäcka gryende transkription tillsammans med immunostaining med anti CBX1 och γH2AX antikroppar.
