Biofilme bacteriano são comunidades de bactérias associadas com superfícies biológicas ou artificiais 1-3. Eles formam por um mecanismo de adesão-crescimento juntamente com a produção de matriz extracelular rica em polissacárido que protege e estabiliza o edifício 4,5. Estes biofilmes não são assemblages simplesmente passivos de células aderidas a superfícies, mas organizado e sistemas biológicos complexos dinâmicos. Quando bactérias mudar de planctônicas de biofilme estilo de vida, as alterações na expressão de genes e fisiologia celular estão observada, bem como um aumento da resistência aos antibióticos e as defesas do hospedeiro imunes estando na origem de muitas infecções persistentes e crónicas 6. No entanto, o desenvolvimento controlada dessas estruturas vivas também oferecem oportunidades para aplicações industriais e ambientais, como a biorremediação de depósitos de resíduos perigosos, bio-filtração de água industrial ou formação de bio-barreiras para proteger o solo e as águas subterrâneas de contaminção. Para avançar no sentido de uma descrição microescala das propriedades físicas do biofilme, as novas ferramentas dedicadas são obrigatórios. Este documento detalha uma abordagem original concebido para alcançarmedição dos parâmetros mecânicos locais in situ, sem perturbar o biofilme e permitindo que o desenho da distribuição espacial das propriedades dos materiais em microescala e, em seguida, a heterogeneidade mecânica. O princípio do ensaio baseia-se na dopagem de um biofilme cresce com micropartículas magnéticas, seguida da sua carga remota usando uma pinça magnéticos no biofilme maduro. Deslocamento de partículas sob controlado aplicação de força magnética fotografada ao microscópio permite derivação parâmetro viscoelástico local, cada partícula relatando seu próprio ambiente local. A partir destes dados, o perfil mecânico 3D do biofilme pode ser desenhada, revelando dependências condições espaciais e ambientais. Aderência bacteriana e crescimento em interfaces levar à formação de estruturas tridimensionais heterogéneos chamados biofilmes. As células que habitam estas estruturas são mantidas juntas por interacções físicas mediadas por uma rede de substâncias poliméricas extracelulares. Biofilme bacteriano impactar muitas atividades humanas ea compreensão de suas propriedades é fundamental para um melhor controle do seu desenvolvimento - manutenção ou erradicação - dependendo de seu resultado adverso ou benéfico. Este artigo descreve uma nova metodologia com o objetivo de medir in situ as propriedades físicas locais do biofilme que tinha sido, até agora, examinada apenas a partir de uma perspectiva macroscópica de material e homogênea. A experiência aqui descrita envolve a introdução de partículas magnéticas num biofilme em crescimento sobre as sementes de sondas locais que podem ser comandadas à distância, sem perturbar as propriedades estruturais do biofilme. Pinças magnéticas Dedicados foram desenvolvido para exercer uma força definida em cada partícula incorporada no biofilme. A instalação é montada sobre a plataforma de um microscópio, para permitir a gravação de imagens de lapso de tempo do período de puxar partícula. As trajectórias das partículas são, em seguida, extraiu-se a partir da sequência de puxar e os parâmetros locais viscoelásticas são derivados a partir de cada curva de deslocamento das partículas, proporcionando assim a distribuição 3D-espacial dos parâmetros. Ganhar insights sobre o perfil mecânico do biofilme é essencial do ponto de vista de um engenheiro de vista para fins de controle do biofilme, mas também a partir de uma perspectiva fundamental para esclarecer a relação entre as propriedades arquitetônicas e biologia específico dessas estruturas.

Uma análise típica irá proporcionar a distribuição espacial dos parâmetros viscoelásticos na escala mícron sobre um biofilme viver sem perturbar o seu arranjo original.
