De beweging van cellen in het lichaam, vaak over grote afstanden, is nodig voor de groei en ontwikkeling, wondgenezing en immuunrespons. Celmigratie is complex en vereist de coördinatie van verschillende processen, waaronder signaal transductie en herschikking van cytoskelet componenten. Cellen kunnen willekeurig evenals bewegen (chemokinesis) richting gedefinieerde chemische gradiënten (chemotaxis). Vele technieken zijn ontwikkeld om celmigratie in vitro bestuderen. De oudste en meest gebruikte techniek, de Boyden-kamer, bestaande uit een verticaal tweekamersysteem waarbij een chemoattractant stof wordt in de onderste kamer en cellen van belang worden geplaatst in de bovenste kamer 1. De beweging van cellen in de permeabele filter met poriën van gedefinieerde grootte, die de twee kamers wordt bewaakt. Aanvullende technieken ontwikkeld om celmigratie te onderzoeken, waaronder Zigmond kamer 2 en kamer Dunn3. Deze collectieve benaderingen hebben aanzienlijke inzicht in de beweging van vele verschillende celtypes opgeleverd. Naast ondervragen de basisprincipes van chemokinesis en chemotaxis, zijn twee kamers assays onderzoek van celmigratie door extracellulaire matrixcomponenten en zowel endotheliale en epitheliale lagen vergemakkelijkt. Een voordeel van twee-kamer systemen via andere technieken is dat het poreuze membraan kan worden bekleed met eiwitten zoals collageen of fibrinogeen, en celmigratie in een extracellulaire matrix-achtige barrière kan worden beoordeeld. Daarnaast kunnen gekweekte cellijnen worden gekweekt en gedifferentieerd op permeabele steunen. Om de beweging van cellen in de endotheliale barrière te onderzoeken, worden gekweekte endotheelcellen gezaaid en gekweekt in het bovenste reservoir van de permeabele steunen. Beweeglijke cellen, zoals immuuncellen, toegevoegd aan het bovenste reservoir en migratie in het onderste reservoir en in dedothelial barrière in de fysiologische apicale-to-basolaterale richting wordt waargenomen. Dit model is van onschatbare waarde geweest bij het begrijpen van extravasatie van immuuncellen uit het bloed. In tegenstelling tot transendotheliale migratie, de beweging van cellen in een epitheliale barrière treedt meestal in de basolaterale naar apicale richting. Om modelleren deze gebeurtenissen in vitro onderzoekers zaad en groeien gekweekte epitheelcellen aan de onderzijde van de permeabele steunen. Om onze understandi verderng aangeboren immuunrespons bij de urinewegen tijdens infectie met uropathogene Escherichia coli (UPEC), ontwikkelden we een in vitro test, de transuroepithelial neutrofielen migratie assay, dat onderzoek van polymorfonucleaire leukocyten maakt (PMN; neutrofielen) beweging over een blaas epitheelbarrière 6 -8. Zoals bij andere twee kamers modellen transepitheliale migratie worden gekweekte menselijke blaas epitheelcellen gegroeid op de onderzijde van een doorlaatbare steun en vormen confluente epitheliale lagen. Menselijke neutrofielen, geïsoleerd uit veneus bloed, worden aangebracht op de basolaterale zijde van de epitheliale laag en migratie over het epitheel van de fysiologisch relevante basolaterale naar apicale richting wordt gekwantificeerd in reactie op infectie door verschillende stammen van E. coli of de aanwezigheid van chemoattractant moleculen. Veel onderzoek was gericht op neutrofielen beweging, zowel chemokinesis en chemotaxis, bij het ontbreken van extra celtypes. De transuroepithelial neutrofielen migratie test is voordelig omdat het rekening houdt met de complexe interacties tussen de blaas epitheel cellen en immuuncellen tijdens infectie. Deze soepele in vitro model heeft het potentieel om de gedetailleerde onderzoek immuunrespons uroepithelial oppervlakken mogelijk.
