1. Fabricage van Parylene Patronen op SiO 2: Process Flow (zie figuur 1) <ol><li> Ontwerp gewenste Parylene-C-configuratie met een lay-out editor software pakket, geschikt voor het lezen / schrijven van CIF (Caltech tussenvorm) of GDS-II (Graphic Database System-II)-bestanden. Onderdompeling van chips in piranha zuur dient niet alleen om eventuele resten van organisch materiaal te verwijderen, maar ook etsen het substraat oppervlakken. Dit is cruciaal voor dat werkelijke activering met foetaal runderserum. Doet u dit niet voorkomt cel-patronen en diepgaand verandert cel gedrag on-chip. Er is geen verplichting om chips te steriliseren na het reinigen met piranha zuur. Inderdaad sterilisatie door UV belichting is aangetoond cel patronen ondermijnen op een dosis-afhankelijke wijze 13. Zorg moet worden genomen om te wassen korting op alle resterende fotolak na de fotolithografische proces. Aanhoudende fotolak kan fungeren als een ongewenste cyto-kleeflaag die patroonvorming gedicteerd door parylene-C/SiO 2 geometrie overschrijft. Aceton is effectief als het gebruik van de fotolithografische bovenstaande proces en met de opgegeven reagentia beschreven. Echter kunnen andere soorten fotoresist een ander oplosmiddel nodig. Om de impact en het succes van de differe beoordelennt fabricagestappen, kan de contacthoek van de twee contrasterende substraten gemeten. Figuur 2 toont de veranderingen die optreden tijdens de chip activeringsproces. Het is echter waarschijnlijk dat specifieke lijm en afstotende eiwitcomponenten in serum uiteindelijk kan de-paryleen gevormde chip om zijn respectieve cyto lijm of cyto-afstotende eigenschappen uitoefenen. Alle representatieve resultaten gebruikt chips met een parylene dikte van 100 nm, al hebben we met succes hebben gevormd met behulp van zowel dikker en dunner parylene lagen. Belangrijk is dat deze fotolithografische ets techniek maakt veel grotere driedimensionale bediening van parylene configuratie dan hier afgebeeld. Dit opent de weg naar het creëren van celculturen met gedefinieerde driedimensionale topografie, die verder gaat dan gewoon dicteren regio&#39;s van cel adhesie / repulsie, in potentie een middel voor de integratie microfluïdische kanalen in het construct. Zoals getoond, maar dit patroon platform niet universeel doeltreffend in celtypen. Verschillende cellijnen, met hun gevarieerde celadhesiemolecuul profielen, niet verwonderlijk anders gedragen wanneer gekweekt op dit platform. We zijn nog niet geïdentificeerd de belangrijkste componenten in het serum, noch de gratis cel-membraan receptoren, die deze cel-patronen platform ondersteunen. Daarvoor in de toekomst belooft het nut en specificiteit te verbreden. Zo kan een &quot;niet-patronen &#39;cellijn genetisch gemodificeerd worden om de vereiste adhesie molecuul expressie bevorderen en aldus patronen.

Begrip mechanismen die celadhesie en patroonvorming dicteren kunststoffen is belangrijk voor toepassingen zoals tissue engineering, drug discovery, en de fabricage van biosensoren 1-3. Vele technieken zijn beschikbaar en evolueren, elk gebruik te maken van de talloze biologische, chemische en fysische factoren die celadhesie beïnvloeden. Hier beschrijven we een cel-patronen techniek die processen in eerste instantie ontwikkeld voor micro-elektronische fabricage doeleinden gebruikt. Als zodanig is het platform is goed geplaatst om de downstream-integratie van micro-elektronische technologieën, zoals MEA staat, in de patroonvorming platform. De interface tussen een celmembraan en een aangrenzende materiaal is bidirectioneel en complex. In vivo, extracellulaire matrixeiwitten zorgen voor structuur en sterkte en effect op celgedrag via interacties met celadhesie receptoren. Ook cellen in vitro interactie met synthetische substraten via geabsorbeerd lagen van eiwitten 4 terwijl fysisch-chemische invloeden ook hechting moduleren. Bijvoorbeeld, een polymeer oppervlak kan meer &quot;bevochtigbaar&quot; (hydrofiele) van ionen of ultraviolet licht bestraling of etsen gemaakt door behandeling met zuur of hydroxide 5. Gevestigde methoden voor mobiele patroonvorming profiteer van deze en andere cel adhesie mediators. Voorbeelden zijn inkjet printen 6, microcontact stempelen 7, fysieke immobilisatie 8, microfluidics 9, real-time manipulatie 10, en selectieve moleculaire assemblage patroonvorming (SMAP) 11. Elk heeft specifieke voordelen en beperkingen. Een belangrijke drijfveer in ons werk, is echter naar cel patronen te integreren met micro-elektromechanische systemen (MEMS). MEMS verwijzen naar zeer kleine mechanische apparaten aangedreven door elektriciteit. Dit overlapt met de nanoschaal equivalent, nanoelectromechnische systemen. Dit concept werd praktisch alleen als halfgeleider strategieën mogelijk fabricage plaatsvinden op de microschaal. Microfabricage technieken oorspronkelijk ontwikkeld voor de halfgeleider elektronica onbedoeld bruikbaar voor andere toepassingen zoals cellulaire elektrofysiologie bijvoorbeeld gevonden. Een belangrijke downstream doel is om dergelijke micro-elektronische technologieën met een high fidelity cel patronen proces (het vormen van een bioMEMS apparaat) te combineren. Verschillende bestaande en anders betrouwbare en praktische cel-patronen technieken verenigbaar zijn met dit idee. Bijvoorbeeld nauwkeurige uitlijning van elke ingebedde micro of biosensoren is fundamenteel voor hun doeltreffendheid maar is zeer moeilijk te bereiken met behulp van een techniek zoals microcontact stempelen. Om dit probleem te omzeilen, zijn we bezig met een SiO 2-gebaseerde patronen platform dat fotolithografisch gedrukt parylene-C gebruikt. Fotolithografie impliceert overdracht van geometrische kenmerken vaneen masker op een substraat via UV belichting. Een masker is ontworpen met behulp van een geschikte computer-aided design programma. Bij een glasplaat, een dunne laag ondoorzichtige chroom vertegenwoordigt de gewenste geometrisch patroon (een kenmerk resolutie van 1-2 mm mogelijk). Het substraat een patroon wordt bedekt met een dunne laag fotolak (een UV-gevoelige polymeer). De beklede polymeer wordt vervolgens uitgelijnd en gebracht in nauw contact met het masker. Een UV bron zodanig dat onbeschermde gebieden bestraald toegepast en derhalve oplosbaar en verwijderbaar te worden in de volgende ontwikkelingsstap waardoor een parylene-C weergave van het maskerpatroon achteren. Dit proces ontstaan ​​tijdens de ontwikkeling van halfgeleiderinrichtingen. Als zodanig zijn silicium wafers vaak gebruikt als substraat. Photolithographic afzetting van parylene-C op SiO 2 is dus een eenvoudige en betrouwbare proces dat routinematig plaatsvindt in micro-elektronische cleanroom faciliteiten. Terwijl parylene heeftverschillende wenselijke biotechniek kenmerken (chemisch inert, niet biologisch afbreekbaar), een beperkende factor zijn directe toepassing in cel patronen is haar aangeboren slechte mobiele hechting, toegeschreven aan zijn extreme hydrofobiciteit. Niettemin Parylene-C is eerder indirect worden gebruikt cel patronen, bijvoorbeeld als afpelbare cellulaire template 12,13. Deze benadering wordt beperkt door slechte resolutie en vereist meerdere stappen. De hier beschreven werkwijze maar gebruikt een zure etsstap, gevolgd door incubatie serum, zodat parylene-C-gebieden worden cel-lijm, door een combinatie van vermindering van hydrofobiciteit en serum eiwitbinding. Het eindresultaat is een constructie uit twee verschillende substraten die, na biologische activatie manifesteren respectieve cyto lijm of cyto-afstotende eigenschappen en dus een doeltreffend cel pattering platform. Belangrijk hoeft biologische ag introducerenouders in de cleanroom faciliteit als de substraten kan worden opgeslagen voor onbepaalde tijd vóór gebruik (waarna ze worden geactiveerd met behulp van foetaal serum of andere activering oplossing). Dit parylene-C/SiO 2 patronen platform is daarom een goede kandidaat voor een coalitie met MEMS componenten, zoals de fabricage processen zo nauw spiegel die gebruikt worden voor micro-elektronische fabricage.

cell patterning on photolithographically defined 
parylene-c: sio2 substrates

Cel patronen platformen ondersteunen brede onderzoeksdoelen, zoals de bouw van voorgedefinieerde in vitro neuronale netwerken en de exploratie van bepaalde centrale aspecten van de cellulaire fysiologie. Naar cel patronen gemakkelijk te combineren met Multi-elektrode arrays (MEA&#39;s) en op basis van silicium &#39;lab op een chip&#39; technologieën, is een microfabrication-compatibele protocol vereist. We beschrijven een methode die afzetting van het polymeer parylene-C op SiO 2 wafels gebruikt. Fotolithografie maakt een nauwkeurige en betrouwbare patroonvorming van parylene-C op micron-niveau resolutie. Na activering door onderdompeling in foetaal runderserum (of een andere specifieke activatie oplossing) resulteert in een substraat waarin gekweekte cellen hechten aan, of respectievelijk afgestoten door, paryleen of SiO2 gebieden. Deze techniek heeft het mogelijk patroonvorming van een breed scala aan celtypen (inclusief primaire murine hippocampale cellen, HEK 293-cellijn, menselijk neuron-achtige teratocarcinoomcellijn, primaire muizen cerebellaire granule cellen en primaire menselijke-glioma afgeleide stam-achtige cellen). Interessant is echter het platform niet universeel, gezien het belang van celspecifieke adhesiemoleculen. Deze cel patronen proces is effectief, betrouwbaar kost, en belangrijker kan in standaard microfabrication worden opgenomen (chip industrie) protocollen, de weg vrijmaakt voor de integratie van micro-elektronische technologie.

De fotolithografische werkwijze van patroonvorming SiO2 met Parylene-C in figuur 1. Na bereiding activering van chips in foetaal runderserum maakt een breed scala aan celtypen worden gevormd in kweek. Onze groep heeft met succes een patroon primaire muizen hippocampuscellen 14-16, de HEK 293 cellijn 17, het menselijk neuron-achtige teratocarcinoom (hNT) cellijn 18, primaire muizen cerebellaire granule cellen en primaire menselijke-glioma afgeleide stam-achtige cellen. Figuur 3 illustreert robuuste patroonvorming van HEK 293-cellen op een parylene patroon bestaande uit ronde knopen met &#39;cross-hair&#39; extensions. Chip activering in dit voorbeeld was met foetaal runderserum. Daarentegen Figuur 4 illustreert het potentieel om de patroonvorming platform te vergroten door alternatieve activering oplossingen. Met een oplossing van runderserumalbumine (3 mg / ml) en<html> fibronectine (1 ug / ml) in HBSS, heeft de vorige patroon Voorschrift geïnverteerd. Figuur 5 illustreert een ander celtype (een primair van mensen afkomstige stam-achtige cellijn afgeleid van een hoogwaardig glioom). Hier, de geometrie van de onderliggende patroon effecten celgedrag, met de in figuur 4A patroon bevordering celproces groei langs dunne parylene-C circuits zoals weergegeven in figuren 4B en 4C. Sommige celtypen niet patroon bij gebruik van de gevestigde foetaal serum activering protocol. Figuur 6 illustreert 3T3 L1 cellen groeien tot confluentie met geen waarneembare cyto-afstotende of cyto-klevende verschil tussen Parylene-C en SiO 2 regio&#39;s. pload/50929/50929fig1.jpg &quot;/&gt; Figuur 1.
