Deze werkwijze toont het gebruik van autonoom bioluminescent zoogdiercellen als in vitro cytotoxiciteit screeningstest waarmee levende cellen continu worden gevolgd gedurende hun leven. Dit protocol is flexibel en kunnen worden aangepast aan specifieke experimentele omstandigheden geschikt als vereist. Bijvoorbeeld, de hier gepresenteerde experiment is geschikt voor het volgen van acute toxische effecten, maar kan worden aangepast aan traag of lange termijn effecten te evalueren door herhaaldelijk beeldvorming ...

In de VS, geneesmiddelen en andere producten voor menselijke consumptie bestemde vereisen uitgebreide evaluatie voordat ze worden goedgekeurd voor gebruik door de consument door de Food and Drug Administration. De financiële last voor het uitvoeren van deze tests is bovenaan de ontwikkelaar 1, die de kosten van de nieuwe compound ontwikkeling en dus aanzienlijk meer vertaalt in een hogere kosten voor de consument. Terwijl traditioneel veel van deze screening heeft proefdieren gebruikt om te fungeren als proxy&#39;s voor de menselijke gastheren, heeft dit bewezen een grote financiële last te zijn, met een geschatte $ 2800000000 jaarlijks besteed aan ADME / Tox (adsorptie, distributie, metabolisme, excretie en toxiciteit ) screening alleen 2 en steeds meer bewijs suggereert dat diermodellen niet betrouwbaar kunnen voorspellen humaantoxicologische reacties 3. Daarom is in vitro humane celkweek-based testing heeft aan populariteit gewonnen in de afgelopen twee decennia vanwege de relatief lagerekosten, hogere doorvoer en betere vertegenwoordiging van menselijke biologische beschikbaarheid en toxicologie 4. Voor celcultuur-toxiciteitstests screeningmethoden verschillende eindpunten, zoals het meten van ATP, het screenen van de activiteit van endogeen beschikbaar cytoplasmatische enzymen, sonderen van de integriteit van het celmembraan, of het bijhouden van het niveau van mitochondriale activiteit, cellulaire viabiliteit 5 , 6. Ongeacht het gekozen eindpunt deze werkwijzen vereisen alle de vernietiging van het monster voor metingen kunnen worden verricht, zodat enige producerende data op een tijdstip. Als gevolg hiervan, grote aantallen monsters moeten worden voorbereid en behandeld in parallel voor basis toxicologische kinetiek studies, opnieuw toe te voegen aan de kosten en de arbeid die nodig is voor de nieuwe compound ontwikkeling. Als alternatief testen met behulp van luciferase uitgescheiden zoals Gaussia luciferase 7, Vargula luciferase 8, en Metridia luciferase 9ontwikkeld die de behoefte aan cellysis elimineren en vereisen een fractie van de media eindpunt meting, maar deze zijn nog steeds beperkt tot bemonstering op vooraf bepaalde punten en ook de toevoeging van exogene substraten activerende licht vereisen. Ten nadele van vereiste monster vernietiging en de kosten van substraten elimineren vermijden is een humane cellijn gemanipuleerd dat drukt de volledige bacteriële bioluminescentie (lux) gen cassette (luxCDABEfrp) om voor continue bewaking van levende cellen die vergelijkbaar aan fluorescente kleurstof-gebaseerde live cell imaging, maar zonder de extra foton-activerende en microscopisch onderzoek procedures. Deze cellijn kan constitutief produceren van een optisch signaal voor continue, directe detectie zonder de noodzaak van externe stimulatie, waardoor vernietiging van het monster te voorkomen. Mechanistisch, de lichtgevende signaal gegenereerd uit deze cellen leidens wanneer de luxAB gevormde luciferase-enzym katalyseert de oxidatie van een langketenige aldehyde (gesynthetiseerd en geregenereerd door de luxCDE genproducten met endogene substraten) in de aanwezigheid van verlaagde riboflavine fosfaat (FMNH 2, die gerecycleerd uit FMN de frp gen product) en moleculaire zuurstof 10. Expressie van de lux-cassette in de gastheercel kan ook licht te produceren en gedetecteerd zonder cellulaire vernietiging of exogeen substraatadditie. Ook de interactie tussen de lux genen en endogeen beschikbare FMN en O2 cosubstrates, en het vereiste van behoud van een omgeving die de omzetting van FMN tot FMNH 2 kan ondersteunen, zodat de resulterende bioluminescente signaal alleen worden waargenomen levende , metabool actieve cellen. Deze eisen zijn eerder misbruikt om aan te tonen dat de lux-based bioluminescerende uitgang sterk correleert met cellulaire bevolking maat 11 en dat toxische verbinding blootstelling schaadt autobioluminescent productie in een dosis-response mode 12. Hier gebruiken we een eerder gekarakteriseerde autobioluminescent menselijke embryonale niercellen (HEK293) cellijn 11 de automatische toxicologische screening van antibiotica van de bleomycine familie met bekende DNA beschadigende activiteit als een representatief voorbeeld van de toepassing van autobioluminescent zoogdiercellen toxiciteit valideren tonen.

Zoogdiercel gebaseerde in vitro assays zijn alom toegepast als alternatieven voor dierproeven voor toxicologische studies, maar zijn beperkt door de hoge monetaire en tijd kosten parallelle monstervoorbereiding die noodzakelijk vanwege het destructieve karakter van vuurvlieg luciferase gebaseerde screeningswerkwijzen . Deze video beschrijft het gebruik van autonoom bioluminescent zoogdiercellen, die niet van de destructieve toevoeging van luciferine substraat als een goedkope en gemakkelijke werkwijze voor het bewaken van de cytotoxische effecten van een verbinding van interesse. Zoogdiercellen stabiel tot expressie volledige bacteriële bioluminescentie (luxCDABEfrp) gencassette autonoom produceren van een optisch signaal dat pieken bij 490 nm zonder toevoeging van een dure en mogelijk storende luciferine substraat excitatie door een externe energiebron, of vernietiging van het monster die traditioneel uitgevoerd bij optische beeldopnames. Deze onafhankelijkheid van externe stimulatie legt de last voor het handhaven van de bioluminescente reactie op de enkele cel, hetgeen betekent dat het resulterende signaal alleen wordt gedetecteerd tijdens actieve stofwisseling. Deze eigenschap maakt het lux-expressie cellijn een uitstekende kandidaat voor gebruik als biosentinel tegen cytotoxische effecten omdat veranderingen in bioluminescent productie wijzen op negatieve effecten op celgroei en metabolisme. Ook de autonome karakter en het ontbreken van de vereiste monster vernietiging toelaat herhaalde beeldvorming van hetzelfde monster in real-time gedurende de periode van blootstelling toxisch en kan over meerdere monsters met behulp van bestaande beeldvormende apparatuur op een geautomatiseerde manier worden uitgevoerd.

In deze studie werden de dynamiek van autobioluminescent HEK293 cellen continu gevolgd gedurende een 24 uurs periode in reactie op antibiotica blootstelling (Figuur 1).
