Många in vitro-observationer i modern cellbiologi har utförts på styva täckglas av glas, ofta belagda med ett tunt skikt av ECM-proteiner eller syntetiska peptider innehållande RGD-sekvensen. Dock en sådan grundläggande odlingssubstrat inte rekapitulera hela fysikalisk-kemiska komplexitet ECM och därmed inte ger en exakt modell för att studera cellulära mechanotransduction processer. För att lösa detta problem föreslår vi ett enkelt alternativ till funktionalisera tvådimensionella hydrogeler med ...

Många aspekter av den lokala cellulära mikromiljön (t.ex. styvhet, porstorlek, typ av proteiner eller cell ligandtäthet) ge en koordinat uppsättning regulatoriska signaler som styr cellulära processer som motilitet, celltillväxt, differentiering och genuttryck. Modifieringar av de fysikalisk-kemiska egenskaperna hos den extracellulära miljön kan uppfattas av celler och orsakar olika fysiologiska konsekvenser, inklusive deformation av cellulär polarisering, migration och differentiering. Det förblir dock oklart hur celler översätter ECM ändringar i cellulära biokemiska signaler. Det är därför av stor vikt att konstruera kontrolleras in vitro mikromiljöer som kan återge samspelet mellan celler och deras mikromiljö för att studera mechanotransduction vägar. För att lösa detta problem har vi nyligen infört en ny metod 1, som kallas hydroxy-PAAM hydrogel, för att enkelt skapa två-dimensional mjuka matriser som tillåter att självständigt styra viktiga mechanotransduction ledtrådar: matrisstyvhet, cellgeometri och inneslutnings, slag av protein och cell ligandtäthet. ECM styr cellulära processer via gradienter i morfogener (kemotaxi), självhäftande proteiner (haptotaxis) och stelhet (durotaxis). Under de senaste decennierna, avancerade in vitro-plattformar har utvecklats för att isolera dessa extracellulära signaler för att locka fram hur celler kan översätta biokemiska och biofysiska funktioner i fysiologiska processer 2-5. Elektronstråle 6, fotolitografi 7, fotokemisk immobilisering 8 eller plasmaassisterade tekniker 9 har utvecklats för att styra tillväxten av levande celler på micropatterned substrat. Bland dessa tekniker, microcontact utskrift (μCP), har framträtt som en robust och tillgänglig metod för att skapa cell självhäftande mikro öar 10. På senare tid har ett omfattande arbete 11-14 gjorts för att utveckla μCP på hydrogeler med sökbara stelheter för att återge de många stelheter som observerats i levande vävnader. Bland dessa verk, har polyakrylamid (PAAM) blivit populära 15 och är redan en av de vanligaste polymerbaserade matriser för cell biomekanik analyser. PAAM ytor är vanligtvis funktionaliserad med den heterobifunktionella tvärbindaren N-sulfosuccinimidyl-6-[4&#39;-azido-2&#39;-nitrophenylamido] (sulfo-SANPAH) och ECM-proteiner är kopplade till ytan genom UV-aktivering av sulfo-SANPAH nitrofenyl azid grupper 16. Hynd och medarbetare infört en teknik för mönstring biomimetiska hydrogelpartiklar ytor med protein och peptider som kräver fotopolymerisation i närvaro av ett acroyl-streptavidin monomeren 18. Mer nyligen, Tseng et al. Har rapporterat en ny micropatterning metod 19 baseras på djup UV-exponering av PAAM genom en optisk kvartsmask som kräver att inkubera aktiverade PA-geler med en-etyl-3-[3-dimetylaminopropyl] karbodiimid-hydroklorid (EDC) och N-hydroxisuccinimid (NHS) vattenlösningar före lägg proteinet. Trots möjligheten för dessa tekniker för att skapa homogena och reproducerbara proteiner micropatterns, de flesta av dem lider stora begränsningar: långa syntesprocesser (t.ex. dialys, frystorkning, etc), dyra kemiska föreningar (t.ex. hyaluronsyra, sulfo-SANPAH) eller djup UV bestrålning. Dessutom har dessa tekniker tillåter inte oberoende modulering av substrat stelhet, micropatterngeometri, ECM-protein natur, och cell-ligandtäthet. Med dessa begränsningar i beaktande, har vi utvecklat en ny och enkel akrylamid baserad metod som gör att immobilisering av en mängd olika proteiner och biomolekyler på mjuka hydrogeler och medger oberoende justering av mechanotransduction ledtrådar för att dechiffrera deras roll på cellulära funktioner. Istället för att behandla PAAM hydrogeler med hårda kemiska föreningar, introducerar vi en kommersiell akrylamidmonomer med hydroxylgrupper under PAAM polymerisation. Denna enkla operation övervinner den inneboende antiklister egendom PAAM hydrogeler utan andra tekniska krav. Närvaron av hydroxylgrupper leder till en hög affinitet för hydroxy-PAAM hydrogel för proteiner och biomolekyler som bildar vätebindande interaktioner. I kombination med μCP, hydroxy-PAAM hydrogeler möjliggör en snabb generation tvådimensionell kultur plattform med en oberoende kontrollpå matrix stelhet, typ av ECM-proteiner, cell ligandtäthet och begränsade vidhäftning, som är tänkt att vara en kraftfull plattform för att studera mechanotransduction. Syftet med detta protokoll är att ge den information som behövs för att enkelt göra hydroxy-PAAM hydrogeler utan kompetens inom materialvetenskap. Det slutliga målet är att tillhandahålla ett sätt för forskare att ställa fysiologiskt relevanta frågor på cellulär och vävnadsnivåer som kan leda till en bättre förståelse av mechanotransduction involverade i patofysiologiska mekanismer.

preparation of hydroxy-paam hydrogels for decoupling the effects of mechanotransduction cues

Det är nu väl etablerat att många cellulära funktioner regleras av interaktioner av celler med fysikalisk-kemiska och mekaniska signaler i deras extracellulära matrisen (ECM) miljö. Eukaryota celler ständigt känna sin lokala mikromiljö genom ytan mechanosensors att omvandla fysiska förändringar av ECM till biokemiska signaler, och integrera dessa signaler för att åstadkomma specifika förändringar i genuttryck. Intressant att fysiokemiska och mekaniska parametrarna hos ECM kan par med varandra reglera cell öde. Därför är en nyckel till förståelsen mechanotransduction är att frikoppla den relativa betydelsen av ECM ledtrådar på cellulära funktioner. Här presenterar vi ett detaljerat försöksprotokoll för att snabbt och enkelt skapa biologiskt relevanta hydrogeler för oberoende inställning av mechanotransduction ledtrådar in vitro. Vi kemiskt modifierade polyakrylamid hydrogeler (PAAM) för att övervinna sin naturligt icke-ADHESive egenskaper genom att införliva hydroxyl-funktionaliserad akrylamidmonomerer under polymerisationen. Vi fick en ny PAAM hydrogel, som kallas hydroxy-PAAM, som medger immobilisering av önskad karaktär ECM-proteiner. Kombinationen av hydroxy-PAAM hydrogeler med microcontact utskrift gör det möjligt att självständigt styra morfologi enkelceller, matrisen styvhet, natur och tätheten av ECM-proteiner. Vi erbjuder en enkel och snabb metod som kan sättas upp i varje biologi labb för att studera i cell mechanotransduction vitro processer. Vi validerar denna roman tvådimensionell plattform genom att utföra experiment på endotelceller som visar en mekanisk koppling mellan ECM styvhet och kärnan.
