Bij onderzoek naar de interactie tussen plant virussen en andere pathogenen met hun aangetaste plant gastheren, is het belangrijk om de pathogenen en hun nucleïnezuren in situ te visualiseren, ongeacht of deze tot negatieve effecten op de gastheer. Dit is zeer belangrijk bij het ​​bestuderen pathogeen verkeer in en tussen plantencellen. In situ lokalisatie van genproducten van het pathogeen is een essentiële stap die andere benaderingen voor het bestuderen van de pathogeniciteit proces aanvult. Veel plantenpathogenen, vooral virussen worden overgebracht door insecten, met complexe en intieme interacties met hun vectoren. Lokalisatie van deze virussen in hun vectoren is belangrijk voor het bestuderen van het pad voor transmissie, en de mogelijke interactieplaatsen in de vector. De overdracht van sommige insecten gevectoriseerd plantenvirussen wordt geholpen door endosymbiotic bacteriën die binnen deze insecten wonen. Om beter te bestuderen van de overdracht van deze plantenvirussen by hun vectoren, is het ook essentieel om de endosymbiotic bacteriën te visualiseren in het algemeen, en die betrokken bij virusoverdracht, in het bijzonder. Colocalization van het virus en endosymbiotic bacteriën dus gewenst voor dat mogelijke relaties tussen deze organismen in hun insectengastheer. Afgezien van virusoverdracht, endosymbiotic bacteriën beïnvloeden de verschillende aspecten van de biologie van insecten, waardoor ruimtelijke lokalisatie van deze bacteriën in insecten is van groot belang en het belang. Yellow leaf curl virus tomaat (TYLCV) (Begomovirus, Geminiviridae) is de belangrijkste virale ziekte complex van gecultiveerde tomaat wereldwijd 1-4. TYLCV is een-floëem beperkte virus en wordt uitsluitend gevectoriseerd door de witte vlieg Bemisia tabaci 5,6. Deze transmissie proces wordt verondersteld te worden gemedieerd door onbekende receptoren die de viruscapside herkennen. TYLCV wordt gedacht aan B. te steken tabaci midgut 6,10, en wordt geabsorbeerd door de primaire speekselklier 6 voordat het wordt geïnjecteerd in de plant. In de wittevlieg hemolymph, TYLCV interageert met een GroEL eiwit geproduceerd door het insect secundaire endosymbiotic bacterie Hamiltonella. Deze interactie zorgt voor veilig transport van TYLCV in de hemolymfe, en beschermt tegen aanvallen van het insect immuunsysteem 11. Hamiltonella en Portiera, de primaire endosymbiont wittevlieg, zijn ondergebracht in bacteriocytes, insectencellen gevonden in de hemolymfe en huis endosymbiotic bacteriën. B. 11 tabaci havens extra endosymbiotic bacteriën zoals Rickettsia, Arsenophonus, Wolbachia, en Fritschea, die kunnen worden gelokaliseerd binnen of buiten de bacteriocytes en hebben verschillende effecten op het insect biologie 12. Verschillende rapporten hebben geprobeerd de lokalisatie van TYLCV in planten en witte vliegen studie door tijdrovende en dure protocollen zoals Transmissie Elektronen Microscopie (TEM), antilichamen en RNA in situ op microscopische dikke gedeelte 10,13, een recente studie beschreef de lokalisatie van plantenvirussen in hun installaties en vector hosts met behulp van een eenvoudig protocol 14. Hier beschrijven we een eenvoudig protocol voor de lokalisatie van TYLCV in B. tabaci ontleed midguts en speekselklieren, en in secties bereid uit TYLCV-geïnfecteerde planten. We de lokalisatie van Portiera de primaire endosymbiont B. beschrijven verder tabaci, en de secundaire endosymbionten Hamitonella, Rickettsia en Arsenophonus. Fluorescentie in situ hybridisatie (FISH) is een naam gegeven aan verschillende technieken gebruikt voor het visualiseren gen transcripten in eukaryotische cellen en kunnen verder worden gemodificeerd om andere componenten in de cel, zoals infectie met virussen en bacteriën te visualiseren. Ruimtelijke lokalisatie en visualisatie van virussen en bacteriën tijdens het infectieproces is een essentiële stap die expressie profiling experimenten zoals microarrays en RNAseq complementair reactie op verschillende stimuli. Inzicht in de tijdruimtelijke infecties met deze middelen een aanvulling op biologische experimenten gericht op het begrijpen van hun interactie met cellulaire componenten. Verscheidene technieken voor het visualiseren virussen en bacteriën zoals reportergen systemen of immunohistochemische werkwijzen zijn tijdrovend en sommige zijn beperkt tot met modelorganismen en vaak complexe methoden. FISH dat RNA of DNA soorten richt in de cel is een relatief eenvoudige en snelle method voor het bestuderen spatiotemporele lokalisatie van genen en voor diagnostische doeleinden.
