Patogeni intracellulari batteriche possono essere concepite come strumenti molecolari per sezionare cascate di segnalazione cellulare a causa della loro capacità di manipolare squisitamente e sovvertire le funzioni cellulari che sono necessarie per l&#39;infezione dei tessuti bersaglio ospitanti. Tra questi batteri patogeni, Listeria monocytogenes è un microrganismo Gram positivo che è stato usato come paradigma per parassitismo intracellulare nella caratterizzazione delle risposte immunitarie cellulari, e che ha avuto un ruolo strumentale nella scoperta di percorsi molecolari che controllano citoscheletro e membrana dinamiche di traffico. In questo articolo descriviamo un saggio microscopica robusto per l&#39;individuazione di fasi tardive infezione cellulare di L. monocytogenes basano sulla marcatura fluorescente di InlC, una proteina batterica secreto che si accumula nel citoplasma delle cellule infette; questo dosaggio può essere accoppiato ad alto throughput schermi piccoli RNA interferenti automatizzati per charziazioni che caratterizzano vie di segnalazione cellulari coinvolti nella up o down-regulation di infezione.
