Neste protocolo de vídeo que demonstrar o método não-invasivo para longitudinal dois fótons de terminações nervosas individuais. Terminações nervosas cutâneas sofrer alterações dinâmicas em diferentes estados fisiopatológicos. As fibras nervosas podem passar pelo processo de degeneração e regeneração ou reestruturação no curso de doenças como neuropatia periférica 2 ou Morton neuroma 3. Após lesão traumática, uma parte importante de terminações nervosas na pele é dinâmica reinervação da área danificada. No entanto, a abordagem comum para a investigação de terminações nervosas é ex vivo cortes histológicos que carece de informações em tempo real sobre os processos em curso 4. Usando marcadores fluorescentes codificados geneticamente, é possível acompanhar as terminações nervosas na pele de animais vivos, obtendo informação rica e significativamente mais relevantes sobre as mudanças estruturais. A investigação de terminações nervosas cutâneas é possível usando microscopia de fluorescência convencional, no entanto, a dispersão de luz forte do tecido da pele prejudica fortemente a qualidadedos dados adquiridos 5. Microscopia multiphoton permite a aquisição de imagens de alta resolução nos tecidos fortemente espalhamento devido à soma não-linear de energia de fótons de luz de excitação, resultando em emissão de fluorescência só do ponto focal da objetiva. Este efeito conduz a um forte aumento da profundidade de penetração e melhoria da relação sinal-ruído para a medição em tecidos da pele 6. Usando o mesmo laser como de imagiologia, é possível produzir dissecção selectiva das fibras nervosas 7. No seguinte protocolo que mostram o método de imagiologia longitudinal de terminações nervosas cutâneas in vivo em ratos transgénicos repórter combinado com lesão selectiva a laser disponíveis no comércio usando sistema multifóton microscópio.

Terminações nervosas na pele estão envolvidas em processos fisiológicos, tais como detecção de 1, bem como em processos patológicos tais como a dor neuropática 2. Seu posicionamento close-to-superfície facilita a imagem microscópica de terminações nervosas da pele na vida animal intacto. Utilizando microscopia multifóton, é possível obter imagens finas superando o problema de dispersão de luz forte do tecido da pele. Repórter ratinhos transgénicos que expressam EYFP sob o controlo do promotor Thy-1 (incluindo os neurónios na periferia neurónios sensoriais) são bem adequados para os estudos longitudinais de terminações nervosas individuais durante longos períodos de tempo até vários meses ou até mesmo ao longo da vida. Além disso, utilizando-se o mesmo laser femtosecond como para a criação de imagens, é possível produzir lesões altamente selectivos de fibras nervosas para os estudos sobre a reestruturação da fibra nervosa. Aqui, apresentamos um protocolo simples e confiável para multiphoton longitudinal in vivo de imagens emicrocirurgia laser baseado no rato terminações nervosas da pele.

Usando a técnica descrita, é possível controlar a mesma fibra após a lesão e para estudar a degradação das terminações nervosas danificadas (Figura 1). Aquisição da pilha com a espessura de 120-150 uM é geralmente adequado para a criação da imagem repetitiva durante vários dias, de modo a manter toda a fibra no campo de visão.
