Neste artigo, é descrito um protocolo para a obtenção de gravações in vivo de atividade neural no peixe-zebra adulto. Métodos de gravação extracelulares são utilizados, proporcionando as medições de tensão de actividade eléctrica dentro de uma pequena região de tecido neural. Este método de investigação envolve monitorizar um grande número de células de um animal comportando 1. electrophysiological recording in the brain of intact adult zebrafish

Anteriormente, estudos eletrofisiológicos em peixe-zebra adulto têm sido limitados a fatia preparações ou para preparativos da Copa do olho e gravações electrorentinogram. Este artigo descreve como um peixe-zebra adulto podem ser imobilizados, entubado, e usado para experimentos in vivo eletrofisiológicos, permitindo a gravação da atividade neural. A imobilização do adulto requer um mecanismo para fornecer oxigênio dissolvido para as brânquias em vez de movimento bucal e opercular. Com a nossa técnica, os animais são imobilizados e perfundidos com água habitat para cumprir este requisito. Uma craniotomia é realizada sob tricaina metanossulfonato (MS-222; tricaina) anestesia para fornecer acesso ao cérebro. O eletrodo primário é então posicionado dentro da janela de craniotomia para gravar a atividade cerebral extracelular. Através da utilização de um sistema de perfusão multitubular, uma variedade de compostos farmacológicos podem ser administrados para os peixes adultos e quaisquer alterações na actividade neuronalpode ser observada. A metodologia permite não só para as observações a serem feitas em relação a mudanças na atividade neurológica, mas também permite que sejam feitas comparações entre larva e adulto zebrafish. Isto dá os investigadores a capacidade para identificar as alterações na actividade neurológica devido à introdução de vários compostos em diferentes etapas da vida.

Este protocolo foi utilizado para medir a actividade neuronal do peixe-zebra adulto in vivo. Estes registros eletrofisiológicos são consistentemente e reproduzível obtida.
