1. Voorbereiding van abdominale ganglia, elektrofysiologische metingen en isolatie van enkele geïdentificeerde neuronen uit abdominale ganglion van Aplysia californica

Het neuron R15 is betrokken bij het reguleren van cardiovasculaire, spijsverterings-, ademhalings- en voortplantingssystemen30. Een regelmatig ritmische barstende activiteit van de AP is een kenmerk van R15. Het menselijk brein is buitengewoon complex met bijna 100 miljard neuronen en biljoenen synaptische verbindingen. Er zijn bijna een gelijk aantal niet-neuronale cellen die interageren met neuronen en hun functie in de hersenen reguleren. Neuronen zijn georganiseerd in circuits die specifiek gedrag reguleren. Ondanks de vooruitgang in ons begrip van hersenfuncties en neurale circuits, is er weinig bekend over de identiteit van circuitcomponenten die een specifiek gedrag controleren. Kennis van de identiteiten van verschillende componenten van een circuit zal ons begrip van zowel cellulaire als moleculaire basis van gedrag aanzienlijk vergemakkelijken en helpen bij het ontwikkelen van nieuwe therapeutische strategieën voor neuropsychiatrische aandoeningen.
De zeeslak Aplysia californica is een werkpaard geweest voor het bepalen van neuronale circuits die ten grondslag liggen aan specifiek gedrag1-14. Het Aplysia zenuwstelsel bevat ongeveer 20.000 neuronen die zijn georganiseerd in 9 verschillende ganglia. De neuronen van Aplysia zijn groot en kunnen gemakkelijk worden geïdentificeerd op basis van hun grootte, elektrische eigenschappen en positie in de ganglia. Aplysia heeft een rijk repertoire aan gedragingen die bestudeerd kunnen worden. Een van de goed bestudeerde gedragingen is de kieuwontwenningsreflex (GWR). De centrale componenten van deze reflex bevinden zich in abdominale ganglia. Onderdelen van de GWR-circuits zijn in kaart gebracht en bijdragen van verschillende componenten zijn bepaald. Belangrijk is dat GWR-circuits associatief en niet-associatief leren ondergaan5,6,15-19. Tientallen jaren van onderzoek naar deze reflex hebben ook verschillende signaleringsroutes geïdentificeerd die een sleutelrol spelen in leren en geheugen20-24.
Verschillende preparaten van Aplysia werden gebruikt om cellulaire en moleculaire basis van geheugenopslag te bestuderen. Deze omvatten het intacte dier2,3, semi-intact preparaat1,7,13,14,16 en reconstitutie van belangrijke componenten van neurale circuits25-29. Een verminderde voorbereiding voor het verkennen van Aplysia ganglia voor de elektrofysiologische en moleculaire analyses van geïdentificeerde neuronale circuits wordt hier beschreven. De volgende vier geïdentificeerde neuronen werden bestudeerd. R15, een barstend neuron, L7 en L11, twee verschillende motorneuronen en R2, een cholinerge neuron werden bestudeerd. R2 is het grootste neuron beschreven in het ongewervelde zenuwstelsel. Kortom, deze methodologie omvat proteasebehandeling van ganglia, elektrofysiologische metingen voor en na farmacologische behandelingen en isolatie van enkele neuronen voor kwantitatieve analyse van genexpressie. Deze methodologie stelt ons in staat om moleculaire analyses te combineren met gelijktijdige opname van meerdere neuronen. Deze methodologie werd met succes gebruikt om reacties van R15- en L7-neuronen op acetylcholine (Ach) te bestuderen door gepaarde intracellulaire opnames. Na elektrofysiologische metingen werden R15 en L7 en andere geïdentificeerde neuronen zoals L11 en R2 geïsoleerd voor kwantitatieve polymerasekettingreactie (qPCR) analyse van expressie van CREB1, een transcriptiefactor die belangrijk is voor geheugenopslag.

Een grote uitdaging in de neurobiologie is het begrijpen van de moleculaire onderbouwing van neurale circuits die een specifiek gedrag bepalen. Zodra de specifieke moleculaire mechanismen zijn geïdentificeerd, kunnen nieuwe therapeutische strategieën worden ontwikkeld om afwijkingen in specifiek gedrag veroorzaakt door degeneratieve ziekten of veroudering van het zenuwstelsel te behandelen. De zeeslak Aplysia californica is zeer geschikt voor het onderzoeken van cellulaire en moleculaire gedragsbasis, omdat neurale circuits die ten grondslag liggen aan een specifiek gedrag gemakkelijk kunnen worden bepaald en de afzonderlijke componenten van het circuit gemakkelijk kunnen worden gemanipuleerd. Deze voordelen van Aplysia hebben geleid tot verschillende fundamentele ontdekkingen van neurobiologie van leren en geheugen. Hier beschrijven we een voorbereiding van het Aplysia zenuwstelsel voor de elektrofysiologische en moleculaire analyses van individuele neuronen. Kortom, ganglion ontleed uit het zenuwstelsel wordt blootgesteld aan protease om de ganglionschede te verwijderen, zodat neuronen worden blootgesteld, maar neuronale activiteit behouden zoals bij het intacte dier. Dit preparaat wordt gebruikt om elektrofysiologische metingen van enkele of meerdere neuronen uit te voeren. Belangrijk is dat na de opname met behulp van een eenvoudige methodologie, de neuronen rechtstreeks van de ganglia kunnen worden geïsoleerd voor genexpressieanalyse. Deze protocollen werden gebruikt om gelijktijdige elektrofysiologische opnames uit te voeren van L7- en R15-neuronen, hun reactie op acetylcholine te bestuderen en de expressie van creb1-gen te kwantificeren in geïsoleerde enkelvoudige L7-, L11-, R15- en R2-neuronen van Aplysia.

De gewichten van dieren die in deze studie werden gebruikt, varieerden van 100-200 g. Volgens de beschreven protocollen voerden we elektrofysiologische metingen en moleculaire analyse uit van neuronen van abdominale ganglia geïsoleerd van dieren variërend van 2-5 g tot 200-300 g.
