PCR kombinert med HRMA er en kraftig teknologi for sebrafisk genotyping. Fordelene med denne fremgangsmåten er dens hastighet, robusthet, og følsomhet for å detektere og med punktmutasjoner. Hele protokoll, fra fin-klippet til smelte-kurve analyse kan utføres på mindre enn åtte timer av et enkelt individ. Sebrafisk (Danio rerio) er et virveldyr modellsystem mye brukt for studier av utvikling og sykdom modellering. Nylig har mange transgene og mutasjons teknologier er utviklet for sebrafisk. Rapid transgenesis teknikker, som regel basert på en Tol2 transposon system 1, har blitt kombinert med forbedrede kloning alternativer for flere DNA fragment montering to. Sink-finger nukleaser (ZFNs) og transkripsjons aktivator lignende effektor nukleaser (Talens) har blitt brukt til å målrette loci i både somatiske og germline celler i sebrafisk 3,4. Disse teknikkene kan effektivt generere genmodifiserte dyr, med høyfrekvente mutasjon etablering og bakterie-nettoverføring 3,4. Til tross for disse fremskrittene, tradisjonelle genotyping teknikker i sebrafisk begrense full effekt av de mutagenese og transgenesis verktøy. PCR etterfulgt av gelelektroforese, noen ganger kombinert med restriction-enzymbehandling, er mye brukt for å detektere genom modifikasjon, men er tidkrevende og mindre følsom for å identifisere små innskudd eller delesjoner. TaqMan probe-analyser har høye initielle kostnader og krever forsiktig optimalisering. Sekvensering av PCR-produkter kan ta flere dager, og er ikke praktisk for storskala screening. Restriction fragment lengde polymorfisme (RFLP) analyse kan bare diskriminere SNPs som påvirker et begrenset utvalg av restriksjonsenzym anerkjennelse sider. Høyoppløselig smelteanalyse (HRMA), et lukket rør post-PCR-analyse-metoden, er en nylig utviklet metode som er rask, sensitiv, billig og mottagelig for screening av store antall prøver. HRMA kan brukes til å oppdage SNPs, mutasjoner og transgener 5-7. Sebrafisk er en kraftig virveldyr modellsystem for å studere utviklingen, modellering sykdom, og utfører medikamentscreening.
