MikroRNA är små icke-kodande RNA som reglerar genuttryck genom att hämma översättning och / eller främja mRNA nedbrytning 2. MicroRNAs tillhör familjen av små (21 till 23 nt i längd) icke-kodande RNA-molekyler, som reglerar genuttryck posttranskriptionellt. microRNAs kan utsöndras i det extracellulära utrymmet där de verkar vara relativt stabil. Medan bestämma förändringar i miRNA uttryck kan vara ett viktigt steg mot att identifiera biomarkörer, utföra miRNA profiler och hantera en stor mängd data kan vara skrämmande. <html> MikroRNA (miRNA) utgör en potent skikt av genreglering genom att styra RISC till målställen belägna på mRNA och följaktligen genom att modulera deras translations repression. Förändringar i miRNA uttryck har visat sig vara involverad i utvecklingen av alla större komplexa sjukdomar. Dessutom visade nya rön att miRNA kan utsöndras till den extracellulära miljön och in i blodomloppet och andra kroppsvätskor där de kan cirkulera med hög stabilitet. Funktionen för sådana cirkulerande miRNA är i stort sett svårfångade, men systematiska hög genomströmning metoder, till exempel miRNA profilering arrayer, har lett till identifiering av miRNA signaturer i flera sjukdomstillstånd, bland annat neurodegenerativa sjukdomar och flera typer av cancer. I detta sammanhang, såsom rapporteras i vår nyligen genomförd studie, som gör identifiering av miRNA expressionsprofil i cerebrospinalvätskan miRNA attraktiva kandidater för biomarkör analys. _content &quot;&gt; Det finns flera verktyg för profilering mikroRNA, som microarrays, kvantitativ realtids-PCR (qPCR), och djupa sekvensering. Här beskriver vi en känslig metod för att profilera mikroRNA i cerebrospinal vätska genom kvantitativ realtids-PCR. Vi använde Exiqon mikroRNA färdiga att använda PCR mänskliga paneler I och II V2.R, vilket möjliggör detektion av 742 unika mänskliga mikroRNA. Vi utförde arrayer i trippel körningar och vi bearbetat och analyserat data med hjälp av Genex Professional 5 programvara. Med användning av detta protokoll har vi framgångsrikt profilerad microRNAs i olika typer av cellinjer och primära celler, CSF, plasma och formalinfixerade paraffininbäddade vävnader.

Resultat från denna studie har tidigare offentliggjorts 1. Figur 1 visar resultat från analysen av kandidatreferens microRNAsna använder geNorm ansökan. Följaktligen två microRNAs, miR-622 och miR-1266, identifierades som referens gener och användes för att normalisera miRNA värden.
