assessing the development of murine plasmacytoid dendritic cells in peyer's patches using adoptive transfer of hematopoietic progenitors

Denne protokollen detaljer en fremgangsmåte til å analysere evnen til renset hematopoetiske stamceller for å generere dendrittiske celler plasmacytoid (PDC) i tarmen Peyer patch (PP). Vanlige dendrittiske cellen stamfedre (CDPs, lin - c-kit lo CD115 + Flt3 +) ble renset fra benmargen av C57BL6 mus ved FACS og overført til mottaker mus som mangler en betydelig PDC befolkningen i PP, i dette tilfellet, IFNAR - / - mus ble benyttet som overføringsmottakere. I noen mus, ble overekspresjon av den dendrittiske cellen vekstfaktor Flt3 ligand (Flt3L) håndheves før adoptiv overføring av CDPs, ved hjelp av hydrodynamisk genoverføring (HGT) av Flt3L-koding plasmid.
