assessing cell cycle progression of neural stem and progenitor cells in the mouse developing brain after genotoxic stress

Serebral korteks nöronlar embriyonik beynin lateral ventriküller astar bir yalancı epiteli oluşturan nöral kök hücrelerin farklı (NSPC), beyin gelişimi sırasında oluşturulur. Gibi iyonize radyasyon gibi genotoksik stresler, NSPC yüksek duyarlılığı ile ilgili gelişen beyin üzerinde son derece zararlı etkileri vardır. Dahil hücresel ve moleküler mekanizmaların açıklanması, hasarlı doku, hücre döngüsü boyunca NSPC ilerlemesi belirlemek için güvenilir bir yöntem gerektirir hücreleri, bu özel tiplerinin DNA hasarı yanıtın karakterizasyonu bağlıdır. Burada hamile farelerde embriyonik ve koronal beyin bölümlerinde bu iki timidin analogları daha saptanmasında edu ve BrdU ardışık intraperitonal enjeksiyonlar göre bir yöntem gösterilmektedir. Edu ve BrdU hem S fazında kopyalayan hücre DNA olarak dahil edilmiştir ve iki farklı teknikler (azid ile tespit edilir ya dabunların aynı anda algılanmasını kolaylaştırmak spesifik bir antikor, sırasıyla). Edu ve BrdU boyama daha sonra dorsal telencephalon standart bir bölgede ventriküler kenar onun mesafesinin bir fonksiyonu olarak her bir NSPC çekirdeği için belirlenir. Bu nedenle bu çift etiketleme tekniği DNA hasarına tepki olarak hücre siklüs hapsinin yol açan bir hücre döngüsü kontrol noktası aktive olması, bu ikinci hücre döngüsü yoluyla ilerleme hücreleri ayırt sağlar.
