In dit artikel presenteren we twee snelle, specifieke en relatief gevoelige colorimetrische microplate assays voor het bepalen van ascorbaat afgeleid van de intra-en extracellulaire compartimenten in gekweekte cellen. De testen kunnen worden aangevuld met toegang tot standaard laboratorium apparatuur en reagentia. De ontdekking van de chemische aard van ascorbinezuur (vitamine C) en de identificatie als het lang gezochte &quot;anti-scheurbuik factor&quot;, door Albert Szent-Györgyi en anderen in papers 1928-1934 1 werden mijlpalen in de geschiedenis biochemie. Inderdaad, deze ontdekkingen hebben bijgedragen aan Szent-Györgyi de toekenning van de Nobelprijs voor de Fysiologie of Geneeskunde in 1937. Hoewel het exacte mechanisme van glutamaat-geïnduceerde ascorbaat vrijlating uit de astrocyten onbekend is, hebben we onlangs bewijs geleverd waaruit blijkt de betrokkenheid van de cel zwelling veroorzaakt door glutamaat opname door de astrocyten glutamaat en aspartaat transporter (GLAST; ook bekend prikkelende aminozuurtransporter isovorm 1 [EAAT1 ] bij de mens) en daaropvolgende activatie van volume-gevoelige osmolyt en anionkanalen (VSOACs) dat doorlaatbaar is voor kleine organische anionen zoals ascorbaat 32 zijn. Bovendien, andere belangrijke factoren beïnvloeden de assay snelheid, gebruiksgemak en toegang tot betrekkelijk gespecialiseerde apparatuur zoals hoge-prestatie vloeistofchromatografie (HPLC) apparaat. Hier presenteren we een eenvoudige en zeer specifieke colorimetrische microplate assay voor de bepaling van intracellulaire ascorbaat in gekweekte cellen, evenals een afzonderlijke assay voor de bepaling van ascorbaat-uitstroming uit gekweekte cellen. De laatste test is bedoeld om het probleem van de onderschatting van ascorbaat vrijlating uit de cellen te omzeilen door snelle heropname van vrijgegeven ascorbaat door natrium-afhankelijke ascorbaat transporters (SVCTs).
