Labellar sensilla varierar i lättheten att inspelning beroende på skillnader i morfologi och anatomisk organisation. Ibland kan en sensillum inte svarar på några tastants, även en som är känd för att framkalla en positiv respons. Den frekvens med vilken detta sker beror på sensillum typen. L sensilla är mest konsekvent lyhörda och är relativt lätta att komma åt på grund av sin längd. I allmänhet S sensilla är genomgående reagerar, men deras korta längd och position på labellum får god kontakt utman...

Den smaks gör en insekt att upptäcka ett stort utbud av lösliga kemikalier och spelar en viktig roll i mottagandet av en näringsrik substans, eller ett avslag på en skadlig eller giftig en. Smaken är också tänkt att spela en roll i mate urval, genom detektion av feromoner 1-5. Dessa viktiga och olika funktioner har gjort det insekts smaken systemet ett övertygande mål för utredning av hur sensoriska system översätter miljö ledtrådar till relevanta beteende utgångar. Den primära enhet av Drosophila melanogaster smak systemet är smaken hår, eller sensillum. Molekyler in i sensillum via en por på sin spets 2,6. Sensilla påträffas på labellum, benen, vingmarginal och svalget 6. På labellum är antalet och placeringen av sensilla stereotypa. Det finns tre morfologiska klasser av sensilla baserat på längd: den långa (L), mellan (I) och korta (S<html> ) Sensilla 7,8. Varje sensillum innehåller antingen två (I-typ) eller fyra (L-och S-typ) smakreceptor neuroner (GRNs) 9. Olika GRNs svarar på olika typer av smak stimuli: bitter, socker, salt och osmolaritet 7,10 och uttrycka olika undergrupper av smakreceptorer 8,11-13. Bara jag och S-typ sensilla innehåller bitter-lyhörd GRNs 8,10. Den GRNs projektet till subesophageal ganglion (SOG) och deras aktivering av smakmolekyler förmedlas till högre centrala nervsystemet för avkodning, vilket resulterar i ett beteende svar 6. Det relativt lilla antalet nervceller och mottaglighet för molekylär och beteendeanalys gör Drosophila smaken systemet en utmärkt modell för utredning av smak system i allmänhet. Den relativa lätthet med vilken systemet kan manipuleras via genetisk mutation eller GAL4-UAS expressionssystem fungerar också som ett värdefullt verktyg 14,15. ontent &quot;&gt; Eftersom dessa sensilla skjuta ut från ytan av den labellum, gör de utmärkta mål för elektrofysiologi. Bränningen av de GRNs kan övervakas med användning av extracellulära inspelning. Historiskt sidoväggen registreringsmetod, som använder en glaselektrod införd i sensillum att spela in neuronal aktivitet, 26 som har använts. Det är dock tekniskt krävande att utföra denna metod, och det är svårt att spela in under längre från varje preparat. Spetsen-inspelningsmetod, som mäter svaret av neuroner med en elektrod som samtidigt levererar en tastant, har sedan dess blivit den metod som före 9,16. Det har använts för att undersöka smaken systemet Drosophila melanogaster 8,10,17,18 samt ett antal andra insektsarter 19-23. Den har i hög grad underlättats genom utvecklingen av tastePROBE förstärkare, som övervann en av de stora nackdelarna med spets-inspelningsmetod genom att kompensera förden stora potentialskillnad mellan referenselektrod och insekts sensillum, vilket gör att GRN aktionspotentialer som skall spelas utan överdriven förstärkning eller filtrering 24. En annan viktig utveckling var användningen av trikolincitrat som inspelnings elektrolyt 25. TCC trycker svaren från osmolariteten känsliga GRN och stimulerar inte saltkänslig referensnumret, vilket svar som genereras av bittra och socker tastants mycket lättare att analysera 25. Här beskriver vi hur spets inspelning av Drosophila labellar sensilla för närvarande utförs i Carlson laboratoriet. Detta protokoll kommer att förklara hur man kan skapa en lämplig elektrofysiologi rigg, hur man förbereder i farten, och hur man utför smak inspelningar. Vi presenterar även några representativa uppgifter som erhållits genom att spela in från delmängder av Drosophila sensilla, samt några vanliga problem och möjliga lösningar som kan uppstå vid användning av dennateknik.

Den perifera smaksvar insekter kan kraftfullt undersökas med elektrofysiologiska metoder. Den metod som beskrivs här gör det möjligt för forskare att mäta smak svar direkt och kvantitativt, vilket speglar den sinnesintryck att insekten nervsystemet får från smak stimuli i sin omgivning. Detta protokoll beskriver alla viktiga steg i att utföra denna teknik. De kritiska stegen i montering ett elektrofysiologi riggen, till exempel val av nödvändig utrustning och en lämplig miljö för inspelning, avgränsas. Figur 5A visar responsen hos en L sensillum till ett socker, sackaros. Samma sensillum reagerar inte en bitter förening visar berberin. Figur 5B som en I-typ sensillum, som innehåller en bitter mottaglig neuron, visar större amplitud spikar som svar på berberin och mindre amplitud spikar som svar på sackaros.
