De intra-cardiale ex vivo injectie protocollen hierboven beschreven zijn ontworpen om hartfunctie gedurende minstens 48 uur in mid-stage (E9.5-E11.5) muizenembryo&#39;s te behouden. Meer dan een decennium geleden, hebben menselijke embryonale stamcellen (HuESCs) is afgeleid van menselijk blastocysts 1. Sindsdien hebben deze cellen het onderwerp van een belangrijk gebied van onderzoek dat onvervulde vragen in ontwikkelingsbiologie mens behandelt te worden. HuESCs zijn verder voorzien van hoop in de regeneratieve geneeskunde. De laatste jaren zijn menselijke geïnduceerde pluripotente stamcellen (iPSCs) gegenereerd van patiënt-specifieke somatische cellen, die modellen van genetische ziekte 2. Vele in vitro protocollen voor differentiatie van embryonale of geïnduceerde pluripotente stamcellen naar verschillende cellijnen, waaronder hart-lijnen 3, zijn gemeld. De gedifferentieerde cellen worden vaak fenotype door analyse van RNA en eiwit expressie, immunokleuring en / of in vitro functionele testen. Echter, pluripotente stamcellen derivaten in een embryonale juiste omgeving worden geplaatst om te testen of ze de cel volledig verwervenl lot van hun embryonale tegenhanger en of ze recapituleren de echte in vivo functie in reactie op de regionale cues. Terwijl de tissue engineering is veelbelovend, maar nog niet voorzien van alle bekende en onbekende signalen van de echte in vivo ontwikkeling van embryonaal weefsel 4,5. Echter, kleurstof en retrovirale cel etikettering zijn meestal toegepast op vroege muizenembryo&#39;s met nog slecht ontwikkelde organen, of kip embryo&#39;s, die beter bereikbaar 8 zijn. Een uitzondering is de hersenen, die gemakkelijker te richten in embryos 9,10. Een dergelijke aanpak is nog niet toegepast op het kloppend embryonale muis hart. Om direct merken vullen met kleurstoffen of virus en lineage tracing in verder gevorderd stadium muizenembryo&#39;s en volwassen muizen uit te voeren, heeft de cel etikettering aanpak gecombineerd met de analyse van transgene muizen met behulp van het Cre / Lox technologie. De Cre / Lox benadering 11 heeft echter een aantal beperkingen in verband met de spatiotemporal specificiteit van de genomische regulatoire gebieden die de expressie van het recombinase drijven, en de efficiëntie van het Cre / Lox recombinatie 12. Bovendien is deze aanpak niet volledig in op de specifieke vragen van de cel-migratie gedreven overname van cel lot als het slechts een voorloper kan labelen na activering van de regulerende gebied gebruikt om Cre expressie rijden. Het kan ook niet van toepassing op menselijke embryo&#39;s voor de hand liggende ethische kwesties. Gezien deze beperkingen we een reeks nieuwe p ontworpenrotocols van verschillende cel merkmiddelen, zoals fluorescerende kleurstoffen, virus, genexpressie modulatoren zoals shRNAs en DNA-gebaseerde mobiele etikettering vectoren of cellen in het muizenembryo op E9.5 en latere ontwikkelingsstadia injecteren in gerichte gebieden van de hart. Het DNA / cel injecties een stereomicroscoop en een eenvoudige micro-injectie toestel gecombineerd met ex vivo embryo cultuur tot 48 uur, of geïsoleerde hart of embryonale explantaatkweek voor 48-72 uur. Bovendien heeft cell lineage tracing studies in de muis na het labelen van cellen met kleurstoffen of retrovirale vectoren meestal beperkt tot vroeg stadium muizenembryo&#39;s met nog slecht ontwikkelde organen gebleven. Om deze beperkingen te overwinnen, hebben we standaard en ultrasoon gemedieerde micro-injectie protocollen aan verschillende agenten te injecteren in gerichte gebieden van het hart in muizenembryo&#39;s op E9.5 en latere stadia van ontwikkeling. Embryonale explant of embryo&#39;s worden vervolgens gekweekt of links om verder te ontwikkelen in de baarmoeder. Deze middelen omvatten fluorescerende kleurstoffen, virus, shRNAs of stamcel-afgeleide stamcellen. Onze aanpak kan voor het behoud van de functies van het orgel terwijl het toezicht op de migratie en het lot van gelabelde en / of geïnjecteerde cellen. Deze technologieën kunnen worden uitgebreid naar andere organen en zal zeer nuttig zijn om de belangrijkste biologische vragen te beantwoorden in de biologie van ontwikkeling.

De injectie protocollen hierboven beschreven, kunnen cellen worden gemerkt en / of geïnjecteerd in de muis embryonale hart. Als proof of concept, zijn verschillende voorbeelden getoond waarin de injectieprotocol en ex vivo AVC explantaat, geïsoleerde hart of geheel embryokweek werden gecombineerd (Figuur 1).
