Er zijn verschillende kritische stappen in het succesvol corneale assay. De eerste maakt uniforme pellets kunnen worden geïmplanteerd en stimulerende vaartuigen. De belangrijkste onderdelen van pellet preparaat zijn 1) met dragervrij groeifactor; 2) te zorgen voor een goede mix van groeifactor met de sucralfaat en Hydron en 3) het verplaatsen van het uiteindelijke mengsel snel maar zorgvuldig uit de eppendorfbuis aan het gaas waar de pellets worden gegoten. Aanbevolen wordt &quot;dummy&quot; pellets worden zonder groei...

Het proces van angiogenese, de vorming van nieuwe bloedvaten te vormen reeds bestaande degenen, is zeer complex en wordt gereguleerd door een aantal endogene factoren die de verschillende stappen van het vaartuig kiemen en morfogenese controleren. Angiogenese wordt veroorzaakt door een verschuiving in balans tussen pro-en anti-angiogene factoren, het evenwicht dat gewoonlijk handhaaft de vasculatuur in een rusttoestand. Angiogenese bij volwassenen voorkomt in bepaalde fysiologische omstandigheden, zoals tijdens de vrouwelijke eierstokcyclus of herstel processen zoals wondheling en weefselregeneratie. Maar het is ook een kenmerk van verscheidene ziekten, waaronder tumoren, auto-immuunziekten en ontstekingsziekten. De betrokkenheid van angiogenese in deze fysiologische en pathologische omstandigheden maakt het een belangrijk onderwerp voor onderzoek en een aantrekkelijk doelwit voor therapie. Vanwege de complexiteit van de angiogenese en de betrokkenheid van verschillende cells en factoren in het proces, zoals endotheelcellen, pericyten, circulerende cellen en stromale cellen in vitro modellen beperkt blijven en kunnen de unieke in vivo micro-omgeving niet herhalen. De belangrijkste in vitro assays van angiogenese zijn grotendeels gericht op het observeren directe effecten op endotheliale cellen en het meten van bepaalde stappen in angiogene proces onder gecontroleerde omstandigheden. Deze testen omvatten het kwantificeren van endotheliale celproliferatie 1, migratie 2, netwerkvorming 3, buisvorming 4 en ontspruit uit bolvormige deeltjes 5. Ex vivo modellen, anders dan in vitro die zijn complexer en bevatten meerdere weefsels celtypes, waarvan een voorbeeld de aorta-ring assay 6. Niettemin, zoals andere systemen, kan het niet de inbreng van circulerende cellen en natuurlijke stroma van endotheelcellen vangen zoals bekend in vivo. Pogingenom angiogenese te studeren onder stroom naar de in vivo omgeving worden uitgevoerd met behulp van microfluïdische systemen 7, echter zelfs deze testen bootsen, hoewel veel verbeterd, zijn nog steeds niet in staat om rekening te houden met alle compartimenten bestaande in vivo. Door de beperkingen van de in vitro en ex vivo angiogenese modellen in vivo modellen blijven betrouwbaarder keuzes voor angiogenese studies. Voorbeelden van deze modellen zijn voorzien van de implantatie van transparante kamers, &quot;ramen&quot;, dat de visualisatie van de groeiende bloedvaten onder de microscoop 8 toestaan, injecteerbare subcutane implantaten zoals matrigel en schip vorming in normale weefsels zoals muis oor en de kip chorioallantoïsmembraan (CAM ). Een van de meest aannemelijk en kwantitatieve in vivo angiogenese modellen de corneale neovascularisatie micropocket assay beschreven, waarbij de natuurlijk avasculaire exploiteerthoornvlies als een &quot;scherm&quot; te visualiseren en evalueren nieuwe angiogene 9 groei. Hier beschrijven we de corneale micropocket assay zoals ontwikkeld in muizen. Aanvankelijk werd het model gebruikt om de niet-specifieke angiogene stimuli bij konijnen hoornvliezen meten. Dit werd gedaan door de invoering van de tumor stukken in de waterige humor van de voorste kamer van het oog van een konijn en gemeten tumor-geïnduceerde neovascularisatie 11. Echter, werd de test later evolueerde naar effecten van specifieke groei bestuderen factoren 10 om beter te specificeren en standaardiseren van de angiogene effect. Om de groeifactor release in het oog werden langzame afgifte pellets bevattende bekende hoeveelheden van angiogene groeifactoren in plaats van weefsels. De beschikbaarheid van gezuiverde recombinante angiogene eiwitten zoals bFGF of VEGF in staat hun gebruik als specifieke doelstellingen van angiogenese modulaters 12. Aanvankelijk was de assayvoornamelijk gebruikt in konijnen, die makkelijker te verwerken vanwege hun grootte, maar later werd het model vertaald in muizen; een kleinere en minder dure diermodel. Verschuift van konijn tot muizen die een belangrijk voordeel van de mogelijkheid om genetisch gemanipuleerde dieren te gebruiken, waardoor een nieuw gebied van onderzoek naar de genetische componenten van invloed zijn angiogenese 13. Naast de meer aanvaardbaar gebruik van corneabepaling bestuderen angiogenese, kunnen andere biologische processen ook onderzocht middels gemodificeerde assays. Zo werden studies lymfangiogenese mogelijk gemaakt door de inplanting van een lage dosis bFGF pellets die de visualisatie van lymfevaten door specifieke moleculaire merkers 14 toegestaan. Daarnaast heeft deze assay een middel om de effecten van straling evalueren angiogenese 15 ontvangen. In sommatie, het hoornvlies micropocket angiogenese assay is een kwantitatieve, reproducible, flexibele evaluatie van de angiogenese in vivo. Een groot voordeel van deze test is dat de meting van achtergrond vaartuigen is nodig omdat de vaten groeien op een natuurlijk avasculaire weefsel. We beschrijven hier het protocol van deze test in de details en bespreken verschillende scenario&#39;s die kunnen optreden. De test bestaat uit 3 afzonderlijke segmenten. Hier zullen we de voorbereiding van de groeifactor-inclusive pellets, de daaropvolgende chirurgische implantatie en tenslotte de methode die wordt gebruikt om de resulterende neovasculaire groei kwantificeren beschrijven.

the corneal micropocket assay: a model of angiogenesis in the mouse eye

De muis cornea micropocket assay is een robuuste en kwantitatieve in vivo analyse voor het evalueren van angiogenese. Via gestandaardiseerde langzame afgifte pellets met specifieke groeifactoren die de groei van bloedvaten leiden gehele natuurlijke avasculaire hoornvlies, kan angiogenese worden gemeten en gekwantificeerd. In deze assay de angiogene respons wordt gegenereerd in de loop van enkele dagen, afhankelijk van het type en de dosering van groeifactor gebruikt. De inductie van neovascularisatie wordt gewoonlijk geactiveerd door hetzij basische fibroblastgroeifactor (bFGF) en vasculaire endotheliale groeifactor (VEGF). Door deze groeifactoren met sucralfaat en Hydron (poly-HEMA (poly (2-hydroxyethylmethacrylaat))) en gieten van het mengsel tot pellets, kunnen ze chirurgisch worden geïmplanteerd in het oog muis. Deze uniforme pellets langzaam laat de groeifactoren over vijf of zes dagen (bFGF of VEGF respectievelijk) waardoor voldoende angiogene respons nodig voor vaartuig gebied quantification met een spleetlamp. Deze test kan worden gebruikt voor verschillende toepassingen, waaronder de evaluatie van angiogene modulator geneesmiddelen of behandelingen en vergelijking tussen verschillende genetische achtergronden invloed angiogenese. Een ervaren onderzoeker na het uitvoeren van deze test kan een pellet implanteren in minder dan 5 minuten per oog.

Typische resultaten voor bFGF en VEGF pellets in de normale lage angiogene C57BL / 6J-muizen zijn getoond in figuur 2A en B, respectievelijk.
