Retinale ganglioncellen (RGC&#39;s) tot expressie L, N, P / Q en T-type VGCC bepaald door farmacologische blokkade van deze onderdelen van de gehele cel Ca kanaalstroom 1,2. Echter, een beperking van het gebruik van dergelijke moleculaire technieken is de vereiste identificatie van de gensequentie. Om eiwitten in het netvlies lokaliseren, immunohistochemie kan worden uitgevoerd op wholemount netvlies. Door de toegankelijkheid van de RGC de wholemountvoorbereiding biedt een uitstekend platform om de lokalisatie van specifieke eiwitten te bestuderen om het RGC somata en hun axonen. Naast hun lokalisatie, kunnen enkele functionele eigenschappen van de VGCCs in RGC worden aangetoond door het gebruik van calcium beeldvormingstechnieken. We beschrijven een calcium imaging protocol selectief labelen RGC met een calcium indicator kleurstof intracellulaire calcium dynamiek meten. Hoewel synthetische calcium indicator kleurstoffen uitstekend platform voor de studie van intracellulaire calcium dynamiek heeft het wijdverbreide gebruik belemmerd door het onvermogen om effectief ladingsspecifiek populaties van neuronen in een bepaald netwerk. Veel technieken zoals bulkbelading 11 en elektroporatie 8,12 uitgevoerd laden gehele celpopulaties zijn dergelijke technieken geen onderscheid tussen specifieke celtypen. Genetisch gecodeerde calcium indicatoren de mogelijkheid om specifieke celpopulaties selectief labelen, maar dergelijke werkwijzen vereisen het genereren van transgene dieren 13. De techniek beschrijft een method om selectief labelen RGC in de wholemount voorbereiding via oogzenuw stomp injectie van een calcium indicator kleurstof. Samen genomen, de structurele en fysiologische technieken die in dit artikel geven een platform om de lokalisatie en de bijdrage van de VGCCs om de calcium signaal in RGC en hun axonen te bestuderen.

In dit artikel beschrijven we de instrumenten, reagentia, en de praktische stappen die nodig zijn voor: 1) succesvol bereiding van wholemount netvliezen voor immunohistochemie en 2) calcium imaging voor de studie van spanningsgevoelige calciumkanalen (VACK) gemedieerde calcium signalering in retinale ganglioncellen. De calcium imaging methode beschrijven we omzeilt kwesties met betrekking tot niet-specifieke belasting van ontheemd amacrine cellen in het ganglion cellaag.
