I retina-gangliecellene (RGCs) uttrykke L, N, P / Q-og T-type VGCC som bestemt ved farmakologisk blokkade av disse Komponentene av hele cellen Ca-kanalstrøm 1,2. Selv om syntetiske kalsium indikatorfargestoffer gir en utmerket plattform for studiet av den intracellulære kalsiumdynamikk, har sin utbredt bruk blitt hindret av manglende evne til å effektivt laste spesifikke populasjoner av nerveceller innenfor et gitt nettverk. Mange teknikker som bulklaste 11 og elektroporering 8,12 er blitt utført for å laste hele grupper av celler, men slike teknikker ikke diskriminerer mellom spesifikke celletyper. Genetisk kodede kalsium indikatorer gir evnen til selektivt å merke spesifikke populasjoner av celler, men slike metoder krever generering av transgene dyr 13. Vår teknikk beskriver en method for selektivt å merke RGCs i wholemount preparat via synsnervestump injeksjon av en kalsiumindikator fargestoff. Tatt sammen, de strukturelle og fysiologiske teknikker beskrevet i denne artikkelen gi en plattform for å studere lokalisering og bidrag av VGCCs til kalsium signal i RGCs og deres aksoner.

immunohistochemical and calcium imaging methods in wholemount rat retina

I denne artikkelen beskriver vi verktøy, reagenser, og de praktiske tiltak som er nødvendig for: 1) vellykket utarbeidelse av wholemount netthinne for immunhistokjemi og, 2) kalsium bildebehandling for studiet av spenning gated kalsium kanal (VGCC) mediert kalsium signalisering i retinal ganglion-celler.
