Omprogrammering mänskliga somatiska celler till hiPSCs rymmer oanade löften i grundläggande biologi, personlig medicin, och transplantation 12. Mänskliga embryonala stamceller (hESCs) har en förmåga att själv förnya in vitro och har pluripotens, vilket kan vara potentiellt användbara för sjukdomsmodellering, för drogscreening, och att utveckla cellbaserade terapier för behandling av sjukdomar och vävnadsskada. Men hESCs har en begränsning för cellersättningsterapi på grund av immunologiska, onkologiska och etiska hinder, samt för att studera sjukdomsrelaterade gener, kan sjukdomsspecifika hESCs isoleras genom preimplantatorisk genetisk diagnostik (PGD) närmar sig, men det är fortfarande tekniskt utmanande och embryodonationer är ganska sällsynta. Dessa frågor är relaterade till framsteg i stamcellsbiologi, vilket har lett till utvecklingen av inducerade pluripotenta stamceller (hiPSCs). Mänskliga iPSCs är genetiskt omprogrammeras från humana vuxna somatiska celler och hamnen pluripotenta stamcellsliknande egenskaper som liknar hESCs, vilket gör dem en användbar källa för regenerativ medicin såsom dmatta upptäckt, modellering sjukdom och cellterapi vid patientspecifikt sätt 1,2. Tills nu, det finns flera metoder för att generera humana iPSCs, inklusive virusmedierad (retrovirus och adenovirus) 3, icke-virusmedierad (BAC Organ och vektorer transfektion) fyra gense transduktioner och proteinleveranssystem 5-7. Även en leverans av virusmedierade gener kan garantera en viss nivå av effektivitet, kan virala vektorer lämna genetiska avtryck, eftersom de integreras i värdkromosomer att uttrycka omprogrammering gener på ett okontrollerat sätt. Även när viral integration av transkriptionsfaktorer kan aktivera eller inaktivera värdgener 8, kan det orsaka en oväntad genetisk avvikelse och risken för tumörbildning 5,9. Å andra sidan, var det direkta införandet av proteiner eller RNA till somatiska celler rapporterats, men har vissa nackdelar, såsom arbetskrävande, upprepade transfektion och låga omprogrammering 7,10. Även episomala och icke-integrerande adenovirus, adeno-associerat virus, och plasmidvektorer är fortfarande relativt hög förbrukning 11. Av dessa skäl är det rimligt att välja icke-integration omprogrammering metoder med hög effekt för iPSC generation och färre genetiska avvikelser. I denna studie använder vi en Sendai-virus baserad omprogrammering. Denna metod är känd för att vara icke-integrerad i värdgenomet och konsekvent producerar mänskliga iPSCs utan transgena integrationer.

För några år sedan, inrättandet av mänskliga inducerade pluripotenta stamceller (iPSCs) inleddes en ny era inom biomedicin. Potentiella användningsområden för mänskliga iPSCs inkluderar modellering patogenes av genetiska sjukdomar, autologa cellterapi efter gen korrigering, och personlig drogscreening genom att tillhandahålla en källa av patientspecifika och symptom relevanta celler. Det finns emellertid flera hinder att övervinna, såsom eliminering av återstående omprogrammering faktor transgen expression efter humana iPSCs produktion.
