Enkele cel gen profilering kan een belangrijk instrument om de functionaliteit van een enkele cel of een gehele populatie te voorspellen zijn. Vanwege technische beperking, heeft hele gen profielanalyse beperkt tot gemiddelden bevolking. Variaties in genexpressiepatronen en niveaus tussen individuele cellen en subpopulaties zijn voorgesteld om onjuiste interpretatie veroorzaken. De meeste hogere eukaryote populaties heterogeen derhalve de analyse van gepoolde populatie, is het vaak moeilijk te interpreteren cellulaire functies. Individuele cellen binnen een populatie kan subtiel verschillend zijn, en deze verschillen kunnen belangrijke gevolgen hebben voor het pand en de functie van de gehele bevolking 1, 2. Vooral menselijke embryonale stamcellen (hESCs) bekend heterogene, die verschillende niveaus van pluripotentie en diverse mogelijkheden veroorzaakt geslacht specificatie fijn specifieke wijze 3, 4 zijn. Zo kunnen verschillende celoppervlak antigenen worden gebruikt om gedifferentieerde pluripotente stamcellen categoriseren, 5 en Austin Smith groep voorgesteld verschillende pluripotentie in muis embryonale stamcellen, op basis van hun morfologie, differentiatie neiging en afhankelijkheid van signaalweg 6. Dit fenomeen werd de hypothese in menselijk embryonic stamcellen 7. Overwegende dat de studies werden uitgevoerd tussen de verschillende stamcellijnen, niet individuele enkele stamcellen, kan het zeer intrigerend zijn om verschillende niveaus van pluripotency analyseren op de enkele cel niveau, die mogelijk invloed heeft op hun differentiatie capaciteiten jegens alle somatische cellijnen. Cellulaire en moleculaire heterogeniteit kan worden gedicteerd door transcriptie profilering, die de &#39;single cell transcriptome&#39; wordt genoemd en benadrukt nieuwe benaderingen voor de kwantificering van genexpressie niveaus 8-10. Voor de analyse van genexpressie niveaus in individuele cellen ontwikkelden we een eenvoudig maar robuust protocol van eencellige kwantitatieve RT-PCR. We bevestigden de effectiviteit en haalbaarheid van ons protocol door het vergelijken van elke helft van enkele cellysaten evenals serieel verdunde totaal RNA van hESC &#39;, wat resulteert in een minimale technische variaties en verschillen. Verder gebruikten we een genetische reporter lijn homogene p isolerenEVOLKING van hESCs gebruik gene targeting systeem. De donor vector voor targeting OCT4 locus (OCT4-2A-EGFP-PGK-Puro construct) en een paar TALEN plasmiden werden gebruikt 11. De donor vector en een paar TALEN plasmiden werden geïntroduceerd in hESCs (H9, WA09) met behulp van onze nucleofectie en stamselectie protocol en het onderhoud van hESC &#39;werd uitgevoerd op basis van onze routine protocol 12. We bevestigden deze genetische reporter lijn uiten EGFP voor OCT4 expressie in OCT4 :: EGFP hESCs. Ons resultaat toont aan dat individuele hESCs (gesorteerd op OCT4 :: EGFP sterk positieve cellen) in het bezit een hoge mate van OCT4 expressie, maar verschillende niveaus van NANOG expressie. Dus moet onze enkele cel genexpressie test nuttig populatie heterogeniteit van pluripotente stamcellen te bestuderen.

Heterogeniteit van stamcellen bevolking belemmert gedetailleerd inzicht in stamcel biologie, zoals hun differentiatie neiging tot verschillende geslachten. Een enkele cel transcriptome test kan een nieuwe benadering voor het ontleden van individuele variatie zijn. We hebben de enkele cel qRT-PCR-methode ontwikkeld, en bevestigde dat deze methode werkt goed in verschillende genexpressie profielen. In enkele cel niveau, elke menselijke embryonale stamcellen, gesorteerd per OCT4 :: EGFP positieve cellen, heeft een hoge expressie in OCT4, maar een ander niveau van NANOG expressie. Onze enkele cel genexpressie test moet nuttig zijn om de bevolking heterogeniteit ondervragen zijn.

Efficiënte en robuuste cel RNA-amplificatie Om de transcriptie variatie tussen hESCs minimaliseren, gebruikten we OCT4 :: EGFP hESC kloon voor FACS zuivering.
