Enkel celle genet profilering kan være et viktig verktøy for å forutsi funksjonaliteten til en enkelt celle eller et hele populasjonen. På grunn av tekniske begrensninger, har hele genet profilering analyse er begrenset til befolkningsgjennomsnitt. Variasjoner i genutrykksmønster og nivåer mellom de enkelte celler og subpopulasjoner har vært foreslått for å forårsake feilaktig tolkning. De høyere eukaryote populasjoner er heterogene og dermed med analyse av sammenslåtte populasjonen, er det ofte vanskelig å tolke deres cellulære funksjoner. Individuelle celler i en populasjon, kan være litt forskjellig, og disse forskjeller kan ha viktige konsekvenser for egenskapen og funksjonen av hele populasjonen 1, 2. Spesielt er humane embryonale stamceller (hESCs) kjent for å være heterogene, noe som fører til ulike nivåer av pluripotency og ulike potensialer til avstamning spesifikasjon i delikat særegne måter tre, fire. For eksempel kan forskjellige celleoverflateantigener anvendes til å kategorisere udifferensierte pluripotente stamceller, 5 og Austin Smith-gruppen foreslått forskjellige nivåer av pluripotency i mus embryonale stamceller, basert på deres morfologi, differensiering og tilbøyelighet til avhengighet av signalveien 6.. Dette fenomenet ble antatt i menneskelige embryonic stamceller 7. Mens de samlede studiene ble utført blant ulike stamcellelinjer, ikke individuelle enkelt stamceller, kan det være svært interessant å analysere ulike nivåer av pluripotency på celle-nivå, noe som potensielt påvirker deres differensiering kapasiteter mot alle somatiske celle slektslinjer. Cellulært og molekylært heterogenitet kan bli diktert av transkripsjon profilering, som kalles &#39;enkelt celle transcriptome&#39; og fremhever nye tilnærminger for å kvantifisere genuttrykk nivåer 8-10. For analyse av genuttrykk nivåer i enkeltceller, har vi utviklet en enkel, men robust protokoll for enkelt celle kvantitativ RT-PCR. Vi bekreftet effekten og gjennomførbarheten av vår protokollen ved å sammenlikne hver halvdel av enkelt cellelysater samt seriefortynnet totale RNA av hESCs, noe som resulterer i minimale tekniske variasjoner og forskjeller. Videre brukte vi en genetisk reporter linje å isolere homogen population av hESCs bruker genet targeting system. Donor vektor for målretting OCT4 locus (OCT4-2A-EGFP-PGK-Puro konstruere) og et par Talen plasmider ble brukt 11. Donor vektor og et par av talen plasmider ble innført i hESCs (H9, WA09) ved bruk av vår nucleofection og klonal valg protokoll og vedlikehold av hESCs ble utført basert på rutinemessig protokoll 12.. Vi bekreftet denne genetiske reporter Line Express EGFP for OCT4 uttrykk i OCT4 :: EGFP hESCs. Vårt resultat viser at enkelte hESCs (sortert etter OCT4 :: EGFP sterkt positive celler) holder høye nivåer av OCT4 uttrykk, men ulike nivåer av NANOG uttrykk. Så, bør vår enkelt celle genuttrykk analysen være nyttig å studere befolknings heterogeniteter av pluripotente stamceller.
