Xenopus laevis extracto de ovo é, um sistema robusto bem caracterizado para estudar a bioquímica de diversos processos celulares. Xenopus extracto de ovo foi utilizada para estudar a rotatividade de proteínas, em muitos contextos celulares, incluindo o ciclo de célula e de transdução de sinal de caminhos 1-3. Aqui, é descrito um método para o isolamento de Xenopus extracto de ovo que foi optimizada para promover a degradação do componente da via Wnt crítico, β-catenina. Dois métodos diferentes são descritas para avaliar a degradação das proteínas β-catenina no Xenopus extracto de ovo. Um método é visualmente informativo ([35 S]-radiomarcado proteínas), enquanto o outro é mais facilmente dimensionado para ensaios de elevado rendimento (firefly proteínas de fusão de luciferase-marcada). As técnicas descritas podem ser utilizadas para, mas não estão limitados a, avaliar retorno da proteína β-catenina e identificar componentes moleculares que contribuem para a sua rotação.
