Lipopolysackarid (LPS) är en cellväggkomponent av gramnegativa bakterier 1. Utmaning till LPS är en väldokumenterad modell av akut lungskada, ett syndrom som kännetecknas av akut neutrofil inflammation och ödem 2. Dessutom är lung neutrofili också ett kännetecken för kronisk obstruktiv lungsjukdom (KOL) 3, och LPS utmaningen hos människa har använts för att modellera KOL exacerbationer 4. Således experimentella modeller av LPS exponering är kliniskt relevanta och värdefulla verktyg för att förstå människans patologi. Syftet med pulmonell leverans av aerosolform LPS beskrivs här är att generera en neutrofil inflammatoriskt svar i ledande och andningsvägarna, utan systemiskt engagemang. Flera tekniker för LPS-utmaning har beskrivits tidigare. Intra-venös injektion av LPS är den mest använda administreringssättet. Även om denna teknik är lättillgänglig, tHan primär skada är att endotelet, med sekundär förstörelse av lung epitel efter neutrofilmigrationen till lungan. Intravenös administrering inducerar även systemisk inflammation 2, vilket kan komplicera den kliniska bilden i djurmodeller. Systemisk inflammation står i kontrast inte observerats med intratrakeal administration. Denna teknik är dock arbetsintensiv och kräver bedövningsmedel samt betydande utbildning 5, 6. Dessutom är lung nedfall genom denna administreringsväg beroende andning 7. Således är lung nedfall påverkas av anestesidjupet behövs för intra trakeal administration och rörlig nedfall i luftvägarna kan observeras. Däremot kräver pulmonell leverans med aerosolform LPS minimal utbildning, och kan lätt åstadkommas på ett stort antal djur med liten eller ingen variation mellan individer 5, 8.En färsk studie bekräftar att aerosoltillförsel är överlägsen den intratrakeal väg när det gäller deponering, och att mer relevanta doser av LPS inducerar neutrofil inflammation med denna modell 8. Tidigare studier har visat att utmaningen till aerosolisPseudoMonas aeruginosa LPS alstrar en markerad inflammatoriskt svar i luftvägslumen och lungparenkym, däribland de alveolära utrymmena 9, 10. Inflammationen karakteriseras av en övervikt av neutrofiler och förekomst av lungödem, och kan således användas för att ta itu med patogenes av akut lungskada och uppnå ytterligare kunskap om de mekanismer som bidrar till sjukdomspatologin.

Akut lungskada (ALI) är en allvarlig sjukdom som kännetecknas av alveolär neutrofili, med begränsade behandlingsalternativ och hög dödlighet. Experimentella modeller av ALI är nyckeln för att öka vår förståelse av sjukdomen patogenes. Lipopolysackarid (LPS) härrörande från grampositiva bakterier inducerar neutrofil inflammation i luftvägarna och lungparenkym av möss. Effektiv pulmonal tillförsel av föreningar, såsom LPS är dock svårt att uppnå. I den metod som beskrivs här, är pulmonell leverans i möss uppnås genom utmaning för aerosoliserad Pseudomonas aeruginosa LPS. Löst LPS aerosol genom en nebulisator kopplad till tryckluft. Möss utsattes för ett kontinuerligt flöde av LPS aerosol i en plexiglaslåda i 10 min, följt av 2 min konditionering efter aerosol avbröts. Utmaning till aerosoliserad P. aeruginosa LPS ger oftast en markerad inflammatorisk reaktion i luftvägslumen och alveolär utrymme, som kännetecknas av en dominans av neutrofiler vid både tidiga och sena tidpunkter.
