Veranderingen in de cel vorm zijn van cruciaal belang in tal van biologische contexten. Bijvoorbeeld, erytrocyten en leukocyten vervormen door capillairen die kleiner zijn dan hun eigen diameter 1. In metastase, moeten kankercellen vervormen door smalle spleten interstitiële alsook kronkelige vaten en lymfatische netwerken om zaad op secundaire locaties 2. Om het fysieke gedrag van individuele cellen te onderzoeken, microfluïdische apparaten presenteren een ideaal platform dat kan worden aangepast aan een reeks van cel gedrag met inbegrip van hun vermogen om te migreren door smalle openingen 3 en passief vervormen door micronschaal vernauwingen 3 9 bestuderen. Polydimethylsiloxaan (PDMS) microfluïdische apparaten zijn optisch transparant, waardoor cel vervormingen worden gevisualiseerd met behulp van een lichtmicroscoop en het gebruik van de basisfuncties voor beeldbewerking gereedschap geanalyseerd. De inrichting is zo ontworpen dat cellen passage door opeenvolgende vernauwingen; deze geometrie is gebruikelijk in fysiologische context, zoals de pulmonaire capillaire bed 12. Naar cel vervormbaarheid peilen, transittijd biedt een handige metric die gemakkelijk wordt gemeten als de tijd die nodig is voor een individuele cel voor doorvoer door een enkele vernauwing 4,6. Om een ​​constante drukval over de vernauwde kanalen tijdens cel doorgang te behouden, maken we gebruik van drukgedreven flow. a microfluidic technique to probe cell deformability

Hier hebben gedetailleerd ontwerp, fabricage en het gebruik van een microfluïdische apparaat om de vervormbaarheid van een groot aantal afzonderlijke cellen te evalueren op een efficiënte wijze. Meestal kan gegevens ~ 10 2 cellen verkregen bij een experiment 1 uur. Een geautomatiseerde beeldanalyse programma maakt efficiënte post-experiment analyse van beeldgegevens, zodat verwerking voltooid binnen enkele uren is. Daarentegen, all-trans retinoic zuur behandelde (neutrofiel-type) HL-60 cellen occluderen de eerste vernauwing slechts 4,3 msec voor passage door de daaropvolgende vernauwingen met een gemiddelde looptijd van 3,3 msec. Deze methode kan inzicht verschaffen in de visco-elastische aard van de cellen, en uiteindelijk onthullen de moleculaire oorsprong van dit gedrag.
