scalable high throughput selection from  phage-displayed synthetic antibody libraries

Efterspørgslen efter antistoffer, der opfylder behovene hos både grundforskning og klinisk forskning applikationer er høj og vil dramatisk stige i fremtiden. Det er dog tydeligt, at de traditionelle monoklonale teknologier er ikke alene op til denne opgave. Dette har ført til udviklingen af ​​alternative metoder til at tilfredsstille efterspørgslen efter høj kvalitet og vedvarende affinitetsreagenser til alle tilgængelige elementer i proteomet. Til den ende har høj kapacitet for udførelse markeringer fra fagpræsenterede syntetiske antistofbiblioteker blevet udtænkt til applikationer, der involverer forskellige antigener og optimeret til hurtig gennemløb og succes. Heri er en protokol, der er beskrevet i detaljer, illustrerer med video demonstration den parallelle udvalg af Fab-fagkloner fra høje mangfoldighed biblioteker mod hundredvis af mål, ved hjælp af enten en manuel 96 kanals flydende handleren eller automatisk robotteknologi system. Ved hjælp af denne protokol, kan en enkelt bruger generere hundredvis af antigener,vælge antistoffer mod dem parallelt og validere antistofbinding inden for 6-8 uger. Fremhævet er: i) en levedygtig antigen format, ii) foreløbige udvælgelse antigen karakterisering, iii) kritiske faser, der påvirker valget af specifikke og høj affinitet kloner, og iv) metoder til udvælgelse overvågning effektivitet og tidlig fase antistof klon karakterisering. Med denne tilgang, har vi fået syntetiske antistoffragmenter (FAB) til mange mål-klasser, herunder single-pass membranreceptorer, udskilte protein hormoner og multi-domæne intracellulære proteiner. Disse fragmenter omdannes let til fuld-længde antistoffer, og er blevet valideret til at udvise høj affinitet og specificitet. Endvidere har de vist sig at være funktionelle i en række standard immunoassays, herunder Western blotting, ELISA, cellulær immunofluorescens, immunopræcipitation og relaterede assays. Denne metode vil fremskynde antistof opdagelse og i sidste ende bringe os tættere på at realisere målet of generere vedvarende, høje antistoffer kvalitet til proteomet.

Den heri beskrevne protokol er blevet anvendt til at isolere antistoffragmenter med en række både structurally- og funktionelt relateret antigen domæner fra kombinatoriske fagpræsenterede Fab biblioteker i parallel.
