Vurdering vandkvaliteten baseret på kemiske og biologiske indikatorer er af afgørende betydning for at få indsigt i de statslige og miljømæssige forhold af en akvatisk økosystem 1-3. Klassiske metoder til kemisk analyse af vand er baseret på organoleptiske egenskaber eller bestemmelse af fysisk-kemiske parametre. Biologiske indikatorer, på den anden side, er dyrearter, hvis tilstedeværelse og levedygtighed give indsigt om de miljømæssige forhold og effekten af ​​forurenende stoffer, for et økosystem, som de opstår i. Typiske eksempler på bioindikatorer er vandlopper en gruppe af små vand krebsdyr, der kan findes i næsten enhver vand levested 4,5. Observere aktivitet og levedygtighed af disse arter fra en vandprøve kan således anvendes til at indhente oplysninger om de generelle betingelser for et økosystem 5.. Larverne af vandlopper som kaldes Nauplii bruge rytmiske slag af deres antenner (hver larve har tre par appendaGES på deres hoved-regionen) at svømme i vand 6. Hyppigheden og intensiteten af disse slag er dermed en direkte indikator for alder, fitness og miljømæssige vilkår for dyret 7-10. Eventuelle undersøgelser på disse prøver er som regel færdig med et mikroskop ved at observere og tælle antenne strøg i Nauplii direkte. På grund af deres størrelse (~ 100-500 um) 11, det kræver ofte at gøre målingerne enten enkeltvis eller at fastsætte en enkelt Nauplius til et substrat. Her demonstrerer vi en ny tilgang til at observere aktiviteten af ​​vandlopper larver i vandprøver ved hjælp af en optisk fanget guld nanopartikel som en ultra-følsom detektor. Optisk pincet anvendes typisk af mange grupper som et fint eksperimentel værktøj til at anvende eller måle kræfter mellem molekyler ned til piconewton området 12-14. For nylig er den vifte af applikationer til optiske pincet blevet udvidet til at observere akustiske vibrationer og løsent udsving i flydende medier ved at overvåge bevægelsen af nano-og mikropartikler, der er indespærret i en optisk fælde 15. Partikler, der er nedsænket i en væske udsættes for Brownsk bevægelse. Inde i en optisk fælde, men denne bevægelse delvist dæmpet af en stærk, laserinduceret gradient kraft. Derfor kan stivheden af ​​optiske fælde og lokalisering af partiklen i fokus laserstrålen tunet af lasereffekten. Samtidig er det muligt at afsløre karakteristika om indfangning potentiale og analysere interaktioner mellem molekyler med partiklen ved at overvåge den tidsafhængige partikel bevægelse i fælden. Denne fremgangsmåde gør det muligt at afhente frekvens, intensitet og retning af den fluide bevægelse, der er genereret af et bevægeligt objekt i flydende miljø. Vi viser, hvordan kan anvendes denne generelle idé at få et frekvensspektrum af bevægelse af en individuel Nauplius uden kravettil direkte forstyrre prøven. Denne eksperimenterende tilgang introducerer et nyt generelt koncept til observation af den bevægelige opførsel af akvatiske eksemplarer i en meget følsom måde. For bemærkninger om bioindicator arter, kan dette udvide den nuværende metode til vandanalyse og kunne anvendes til at få information om sundhed og integriteten af ​​akvatiske økosystemer.

Vi viser, hvordan optiske pincet kan give en følsom redskab til at analysere de strømningstekniske vibrationer frembragt ved bevægelsen af ​​små vandlevende organismer.
