Bovendien wordt electrospinning traditioneel gedaan in een vluchtig oplosmiddel, waarbij de levensvatbaarheid van het eiwit 28 kan beïnvloeden, handhaaft derhalve bioactiviteit van het eiwit worden onderzocht. Deze aanpak specifiek over de combinatie van mechanische, topografische, chemische en lijm signalen naar een afstembare steiger voor perifere zenuw groei te creëren. Steiger monteurs wordt nauwkeurig gecontroleerd door het synthetiserengemethacryleerde Hyaluronzuur (HA). De methacrylation sites worden gebruikt om foto reactieve verknopers hechten. Het verknoopte materiaal niet meer water oplosbare en uitsluitend afgebroken door enzymen 29. De hoeveelheid verknoping verandert de afbraaksnelheid, mechanica en andere fysische eigenschappen van het materiaal. Met HA met 30% methacrylation, die een trekmodulus van -500 Pa heeft, is zacht substraat dat vlakbij de natieve mechanica van neuraal weefsel en kenmerkend voorkeur van neuronen 26,29. Elektrospinnen op een roterende doorn wordt gebruikt om uitgelijnde vezels te creëren voor een topografische cue. Met behulp elektrospinproces samen met microsferen biedt chemische signalen binnen het schavot gedurende langere perioden. Unlike meeste electrospun materialen HA oplosbaar in water zodat het NGF geen agressieve oplosmiddelen tegenkomt tijdens de productie. De emulsie wordt gestabiliseerd met een surfactant, polyvinylalcohol (PVA). De binnenste waterfase bevat het eiwit. Als het wordt toegevoegd aan de oliefase, die de PLGA schaalmateriaal opgelost in dichloormethaan (DCM), de surfactant creëert een barrière tussen de fasen beschermen het eiwit uit de DCM. Deze emulsie wordt dan gedispergeerd in een waterfase bevattende PVA aan het buitenoppervlak van de microsferen te creëren. De stabiele emulsie wordt geroerd om het DCM verdampen. Zodra de productie voltooid is, moet het eiwit worden getest om de levensvatbaarheid verifiëren. Om dit te doen, kan een primaire cel die reageert op NGF worden gebruikt. Dit protocol maakt gebruik van de ganglia (DRG) 8-10 dagen oude kippenembryo&#39;s. De cel bundels worden gezaaid op steigers microsferen gevuld met NGF of als ze leeg zijn. Als NGF nog steeds levensvatbaar zou u verbeterde neuriet groei bij de NGF bevattende scaffolds bekijken. Als NGF niet langer levensvatbaar zal hetniet neurites te breiden bevorderen en moeten vergelijkbaar zijn met de controle verschijnen. De hierin beschreven exacte procedure is gericht op neurale steun echter met eenvoudige wijzigingen van het materiaal, electrospinning methode en eiwitten het systeem kan worden geoptimaliseerd voor verschillende types cellen en weefsels.

electrospinning growth factor releasing microspheres into fibrous scaffolds

This procedure describes a method to fabricate a multifaceted substrate to direct nerve cell growth. This system incorporates mechanical, topographical, adhesive and chemical signals. Mechanical properties are controlled by the type of material used to fabricate the electrospun fibers. In this protocol we use 30% methacrylated Hyaluronic Acid (HA), which has a tensile modulus of ~500 Pa, to produce a soft fibrous scaffold. Electrospinning on to a rotating mandrel produces aligned fibers to create a topographical cue. Adhesion is achieved by coating the scaffold with fibronectin. The primary challenge addressed herein is providing a chemical signal throughout the depth of the scaffold for extended periods. This procedure describes fabricating&#160;poly(lactic-co-glycolic acid) (PLGA) microspheres that contain Nerve Growth Factor (NGF) and directly impregnating the scaffold with these microspheres during the electrospinning process. Due to the harsh production environment, including high sheer forces and electrical charges, protein viability is measured after production. The system provides protein release for over 60 days and has been shown to promote primary nerve cell growth.
