In bloeiende planten, is de somatische-to-voortplantingscel lot overgang gekenmerkt door de specificatie van spore moeder cellen (SMC) in floral organen van de volwassen plant. De vrouwelijke SMC (megaspore moeder cel, MMC) onderscheidt in het eitje primordium en ondergaat meiose. De geselecteerde haploïde megaspore ondergaat mitose aan de meercellige vrouwelijke gametofyt, die aanleiding zal geven aan de gameten, de eicel en de centrale cel te vormen, samen met accessoire cellen. De beperkte toegankelijkheid van de MMC, meiocyte en vrouwelijke gametofyt in de eicel is technisch uitdagend voor cytologische en cytogenetische analyses op enkel cel niveau. In het bijzonder wordt direct of indirect immunodetectie van cellulaire of nucleaire epitopen aangetast door slechte penetratie van de reagentia in de plantencel en single-cell imaging wordt demised door het gebrek aan optische helderheid geheel-mount weefsels. Aldus wordt een efficiënte manier de Nucl analyseren ontwikkelden weoor organisatie en chromatine modificatie op hoge resolutie van enkele cel in whole-mount ingebed Arabidopsis eitjes. Het is gebaseerd op dissectie en inbedding van vaste eicellen in een dunne laag acrylamide gel op een microscopisch preparaat. De ingesloten eitjes worden onderworpen aan chemische en enzymatische behandelingen ter verbetering weefsel duidelijkheid en permeabiliteit voor de immunokleuring reagentia. Die behandelingen behouden cellulaire en chromatine organisatie, DNA en eiwitten epitopen. De monsters kunnen worden gebruikt voor verschillende stroomafwaartse cytologische analyse, waaronder chromatine immunokleuring, fluorescentie in situ hybridisatie (FISH), en DNA kleuring voor heterochromatine analyse. Confocale laser scanning microscopie (CLSM) beeldvorming met hoge resolutie, gevolgd door 3D reconstructie maakt kwantitatieve metingen bij eencellige resolutie.

Wij bieden een robuust protocol voor grootschalige bereiding en verwerking van Arabidopsis eicellen geschikt zijn voor cytologische kleuring in whole-mount. Door de inbedding, de zaadknoppen behouden van een 3-dimensionale structuur (Figuur 3).
