I blommande växter, är det kvinnliga reproduktiva härstamning omgiven av flera cellager inklusive nucellus och fröämnet teguments, vilket gör cytologisk färgning i hela montering tekniskt utmanande. I blommande växter, inrättandet av reproduktiva linjer börjar med differentieringen av SMC, kvinnliga MMC och manliga mikrospor modercell. MMC utvecklas från en sub-epidermal nucellar cell vid den distala spetsen av ovule primordium och mikrospor modercellen utvecklas från sporogena vävnad i ståndarknapp locule, som ligger djupt inne i floral organ 1. SMC genomgå meios för att producera haploida sporer, som sedan ger upphov till gametofyter vid mitos. Den kvinnliga gametophyte eller embryo sac, består av en äggcell, en central cell, två synergids och tre antipodals. Den manliga gametophyte, eller pollen, består av en vegetativ cell och två spermier. Medan den manliga gametophyte fortfarande en relativt tillgänglig objekt av-studien, är den kvinnliga gametophyte inbäddade inuti ovule, själva inneslutna i blomman pistill, och därmed innebär särskilda utmaningar för molekylär och cytologiska analyser. Nyligen dock laser-assistedmicrodissection erbjuds en elegant lösning som gör det möjligt transcriptomic analyser i MMC och kvinnliga gametophytic celler 2-4. Utöver kandidat genuttryck analyser, med hjälp av t ex RNA in situ hybridisering eller reporter gen analyser, cytologiska analyser låter undersöka dynamiken i endogena cellulära komponenter med specifika direkta cellulär färgning eller indirekt immunfärgning. Särskilt cytogenetisk färgning använder FISH och DNA-färgning, tillsammans med immunfärgning av kromatin ändringar eller kromatin komponenter är centrala metoder för att belysa kromatin dynamik och kärnorganisation i Arabidopsis 5. Vanligtvis innebär meios specifika kromosomdynamik som har varit väl undersökta i växt manliga meiocytes 6,7; ytterligare storskaliga, cellspecifika kromatin omorganisation, troligen avspeglar dynamisk epigenetiska omprogrammeringen har beskrivits under pollenutvecklingen 8-10. Däremot, på grund avtill den relativa otillgänglighet av den kvinnliga meiocyte och gametophyte, dessa undersökningar förblir tekniskt svårt att tillämpa, och ofta kräver sektione eller manuell dissektion och enzymatisk nedbrytning (se nedan). Dessutom, den utbredda bristen på optisk klarhet i hela-fäste är ett hinder för högupplöst avbildning av könsceller i intakta fröämnen. En klassisk metod för cytologisk analys av kromosomorganisation i hela-mount fröämnen använder Feulgen s färgning 11-13. Det innefattar syrahydrolys (med användning av underklorsyrlighet) av DNA som resulterar i proteindenaturering och därigenom orsakar förstörelse av kromatinstrukturen. Alternativt kan observeras kromosomorganisation i kvinnliga meiocytes och gametophytic celler använder DAPI färgning och immunfärgning på semi-tunna sektioner eller dissekeras embryo säckar och MMC (till exempel se 14-18). Det är emellertid uppenbart, manuell dissektion och sektionering kan vara arbetsintensiv ochhindrar på kvalitativ och kvantitativ analys av ett stort antal kromatin epitoper. Här ger vi ett effektivt protokoll för att förbereda ett stort antal Arabidopsis fröämnen som lämpar sig för en mängd olika nedströms cytologisk färgning i hela-fäste. I korthet, är blomknoppar inkuberas i en fixeringslösning, är rader av fröämnen dissekeras från pistill och inbäddade i akrylamid på bilden som gjort för pollen meiocytes 19,20. De inbäddade fröämnen vidare rensas och fixerades i metanol, etanol, och xylen före cellväggen matsmältningen och permeabilisering. Möjliga variationer av dessa steg diskuteras. Proverna kan därefter användas för DNA-färgning, immunfärgning, och FISH. Läget preparatet är effektivt och möjliggör parallell försöksuppställningen (upp till 16 diabilder kan beredas i en dag för olika nedströms analys). De behandlingar som beskrivs möjliggör homogena signaler i hela-fäste och välbevarade histologiska, cellulär,och nukleära organisation i könsceller och omgivande nucellar celler som gynnar kvalitativa och kvantitativa jämförelser mellan celltyper. Kalibrerad, CLSM baserade högupplöst avbildning följt av 3-dimensionella rekonstruktion möjliggör meningsfulla kvantitativa mätningar av fluorescenssignaler. Vi använde med framgång denna procedur för att analysera kromatin dynamik i differentiering MMC 21 och utveckla kvinnliga gametophyte 22; Vi presenterar här representativa resultat heterochromatin analys, kromatin immunfärgning, GFP immunfärgning och FISH helt monterade fröämnen. Vi tror också att våra protokoll kommer att vara lämpliga för andra växtvävnader och arter.

an efficient method for quantitative, single-cell analysis of chromatin modification and nuclear architecture in whole-mount ovules in arabidopsis

I blommande växter, är det somatiska-till-reproduktiva cell öde övergång markeras av specifikationen av spor moderceller (SMC) i blom organ i den vuxna växten. Den kvinnliga SMC (megaspore modercell, MMC) skiljer i fröämnet primordium och genomgår meios. Den valda haploid megaspore genomgår sedan mitos att bilda flercelliga kvinnliga gametophyte, vilket kommer att ge upphov till könsceller, äggcellen och central cell, tillsammans med hjälpceller. Den begränsade tillgängligheten av MMC, meiocyte och kvinnliga gametophyte inuti ovule är tekniskt utmanande för cytologisk och cytogenetiska analyser på enskild cellnivå. Speciellt, är direkt eller indirekt immunodetection av cellulära eller nukleära epitoper försämras av dålig penetrering av reagenserna inne i växtcellen och encelliga avbildning demised av bristen på optisk klarhet i hela monterade vävnader. Därför utvecklade vi en effektiv metod för att analysera Nuclöron organisation och kromatin modifiering med hög upplösning av enskild cell i hela monterade inbäddade Arabidopsis fröämnen. Den är baserad på dissekering och inbäddning av fasta fröämnen i ett tunt skikt av akrylamidgel på ett objektglas. De inbäddade fröämnen utsätts för kemiska och enzymatiska behandlingar som syftar till att förbättra tydligheten vävnad och permeabilitet för immunfärgning reagenser. Dessa behandlingar bevara cellulära och kromatin organisation, DNA-och protein epitoper. De prover som kan användas för olika nedströms cytologiska analyser, inklusive kromatin immunofärgning, fluorescens in situ hybridisering (FISH), och DNA-färgning för heterokromatin analys. Confocal laserskanning mikroskopi (CLSM) imaging, med hög upplösning, följt av 3D-rekonstruktion möjliggör kvantitativa mätningar vid encelliga upplösning.

Vi erbjuder en robust protokoll för storskalig bearbetning och beredning av Arabidopsis fröämnen lämpliga för cytologisk färgning i hela-fäste. Tack vare inbäddningen de fröämnen bibehåller en 3-dimensionell struktur (fig 3).
