Dit protocol beschrijft een eenvoudige methode om het beeld automatisch weefsel-specifieke fluorescentie in zebravis embryo&#39;s. Het protocol is ontwikkeld met behulp van een Zeiss Axio Observer. Z1 met Zen Blue 2011-software, maar de techniek kan worden aangepast met behulp van een omgekeerde microscoop met een gemotoriseerde podium en microscoop controle software die tegels kunnen voeren maken van samengestelde afbeeldingen. Transgene zebravis ontwikkeld die het mogelijk maken de directe visualisatie van de activiteit in signaaltransductiewegen zoals fibroblast groeifactoren 1, retinoïnezuur 2 en 3 oestrogenen, in levende embryo. Dergelijke tools kunnen screenen op stoffen die signalerende wegen verstoren (getest als verandering in de fluorescentie-intensiteit) of voor chemische stoffen die moduleren signalering in een weefsel-specifieke wijze (verandering in de fluorescentie lokalisatie) 4. Geautomatiseerde beeldopname verhoogt de doorvoer van chemische schermen dramatisch 5,6. Schermen die automatisch assay fluorescentie in levende zebravis vaak een beroep op dure, gespecialiseerde apparatuur. Zogenaamde high content levert het voordeel van hoge resolutie kwantitatieve beeldvorming maar ten koste van het gebruik van gespecialiseerde plaatlezers ingericht voor confocale microscopie 7,8. Het doel van deze methode is automatisch assay weefsel-specifieke fluorescentie in zebravis embryoin een 96-wells plaat met een standaard epifluorescentiemicroscoop. Weefsel-specifieke fluorescentie kan worden onderscheiden met behulp van deze techniek en kan een redelijke benadering voor laboratoria die de toegang tot gespecialiseerde plaat lezers of high content apparatuur ontbreekt zijn. In dit protocol gebruiken we geautomatiseerde beeldvorming om weefselspecifieke oestrogeenreceptor (ER) agonisten in levende zebravis detecteren 3 dagen na de bevruchting (dpf). De transgene lijn Tg (5xERE: GFP) c262/c262 bevat 5 tandem oestrogeen respons element DNA-sequenties (ERE) stroomopwaarts van groen fluorescerend eiwit (GFP) 3. Bij afwezigheid van ligand, ERs gewoonlijk inactief. Ligandbinding veroorzaakt een conformationele verandering waardoor receptoren ERE DNA te binden en transcriptie te reguleren 9. 5xERE: GFP vis kan worden gebruikt om chemische bibliotheken te screenen ER modulatoren en kan worden gebruikt om de milieu watermonsters oestrogene verontreinigingen screenen.

Zebravis embryo&#39;s zijn een krachtig hulpmiddel voor grootschalige screening van kleine moleculen. Transgene zebravis dat fluorescente reporter eiwitten tot expressie worden vaak gebruikt om chemicaliën die genexpressie moduleren. Chemische schermen die fluorescentie assay in levende zebravis vaak een beroep op dure, gespecialiseerde apparatuur voor hoge gehalte screening. We beschrijven een procedure met behulp van een standaard epifluorescentiemicroscoop met een gemotoriseerde podium om het beeld automatisch zebravis embryo&#39;s en detecteren weefsel-specifieke fluorescentie. Met behulp van transgene zebravis dat oestrogeen receptor-activiteit via expressie van GFP melden, we een semi-geautomatiseerde procedure ontwikkeld voor het screenen op oestrogeen receptor liganden die de verslaggever in een weefsel-specifieke wijze te activeren. In deze video beschrijven we procedures arraying zebravis embryo 24-48 uur na de bevruchting (HPF) in een 96-well plaat en het toevoegen van kleine moleculen die oestrogeenreceptoren binden. Op 72-96 HPF, beelden van elk putje vande hele plaat worden automatisch verzameld en handmatig gecontroleerd op weefsel-specifieke fluorescentie. Dit protocol toont het vermogen aan oestrogenen die receptoren in hartkleppen inschakelen, lever detecteren.

Figuur 1 toont samengestelde beelden van afzonderlijke putjes van een 96-wells plaat. Elk beeld composiet bestaat uit 59 afzonderlijke foto&#39;s met 5% afbeelding overlap. Merk op dat de levende zebravis willekeurig georiënteerd zijn in elk putje, maar we kunnen onderscheiden fluorescentie in het hart van de lever. Helderveld beelden zijn bruikbaar als verwijzingen naar de zebravis oriëntatie te beoordelen en te visualiseren morfologische afwijkingen (figuren 1A en 1C). Geautomatisee...
