Denne protokollen beskriver en grei metode for å automatisk bilde vev-spesifikk fluorescens i sebrafisk embryoer. Protokollen ble utviklet ved hjelp av en Zeiss Axio Observer. Transgen sebrafisk har blitt utviklet som tillater den direkte visualisering av aktivitet i signalveier, for eksempel fibroblast vekstfaktorer 1, retinsyre 2 og østrogener 3, levende embryoer. Slike muliggjør screening for kjemikalier som forurolige signalveier (analysert som endring i fluorescens-intensitet) eller på grunn av kjemikalier som modulerer signalering i en vev-spesifikk måte (endring i fluorescens lokalisering) 4.. Automatisert bildeopptak øker gjennomstrømningen av kjemiske skjermer dramatisk 5,6. Skjermer som automatisk analysen fluorescens i levende sebrafisk ofte er avhengige av dyre, spesialisert utstyr. Såkalt høy-innhold screening gir fordelen av høy oppløsning, kvantitativ bildebehandling, men på bekostning av å bruke spesialiserte plate lesere utstyrt for konfokalmikroskopi 7,8. Målet med denne metoden er å automatisk analyse vev-spesifikk fluorescens i sebrafisk embryoi en 96-brønns plate ved hjelp av en standard epifluorescence mikroskop. Tissue-spesifikk fluorescens kan skilles ved hjelp av denne teknikken, og kan være en rimelig tilnærming for laboratorier som ikke har tilgang til spesialiserte platelesere eller høyt innhold screening utstyr. I denne protokollen, bruker vi automatisert å påvise vevsspesifikk østrogen reseptor (ER) agonister i levende sebrafisk på tre dager etter befruktning (DPF). Den transgene linjen Tg (5xERE: GFP) c262/c262 inneholder fem tandem østrogen respons element DNA-sekvenser (ERE) oppstrøms av grønt fluorescerende protein (GFP) tre. I fravær av ligand, grunnleggende krav er normalt inaktive. Sebrafisk embryo er et kraftig verktøy for storskala screening av små molekyler. Transgen sebrafisk som uttrykker fluorescerende reporter proteiner er ofte brukt for å identifisere kjemikalier som modulerer genuttrykk. Kjemiske skjermer som analysen fluorescens i levende sebrafisk er ofte avhengige av dyre, spesialutstyr for høyt innhold screening. Vi beskriver en prosedyre ved hjelp av en standard epifluorescence mikroskop med en motorisert scene til automatisk bildesebrafisk embryo og oppdage vev-spesifikk fluorescens. Ved hjelp av transgene sebrafisk som rapporterer østrogen reseptor aktivitet via uttrykk for GFP, utviklet vi en semi-automatisert prosedyre for å screene for østrogen reseptorligander som aktiverer reporteren i en vev-spesifikk måte. I denne video beskrives fremgangsmåter for arraying sebrafisk embryoer ved 24-48 timer etter befruktning (HPF) i en 96-brønns plate og tilsette små molekyler som binder østrogenreseptorer. På 72-96 HPF, bilder av hver brønn frahele platen summeres automatisk og manuelt kontrollert for vev-spesifikk fluorescens. Denne protokollen demonstrerer evnen til å oppdage østrogener som aktiverer reseptorer i hjerteklaffene, men ikke i leveren.

Figur 1 viser sammensatte bilder fra individuelle brønner i en 96-brønns plate. Hver sammensatt bilde er sammensatt av 59 enkeltbilder med 5% bilde overlapper hverandre. Legg merke til at den levende sebrafisk er orientert tilfeldig innenfor hver brønn, men vi er i stand til å skille fluorescens i hjertet fra leveren.
