As células estaminais têm a capacidade de se replicar sem limite e o potencial para se diferenciar em qualquer tipo de célula somática. O desenvolvimento de técnicas para reprogramar células somáticas em células-tronco pluripotentes suscitou grande entusiasmo na comunidade científica e entre os médicos, como o advento da regeneração do tecido personalizado está no horizonte 1. Induzido por células-tronco pluripotentes (iPS) apresentam as mesmas características do potencial replicativo ilimitado e pluripotência como células-tronco embrionárias (ES), contornando os dilemas éticos associados à CES. microrna expression profiles of human ips cells, retinal pigment epithelium derived from ips, and fetal retinal pigment epithelium

O objetivo deste relato é descrever os protocolos para comparar o microRNA (miRNA) perfis de humana induzida por células-tronco pluripotentes (iPS), epitélio pigmentar da retina (RPE) derivadas de células iPS humanas (iPS-RPE) e RPE fetal. Os protocolos incluem a recolha de RNA para análise de microarray, e a análise de dados de microarrays para identificar miRNAs que são diferencialmente expressos entre os três tipos de células. Os métodos para cultura de células iPS e RPE fetal são explicados. O protocolo utilizado para a diferenciação de RPE de iPS humana também é descrito. A técnica de extracção de RNA descrevemos foi seleccionado para permitir a recuperação máxima de ARN muito pequena para ser utilizado em um microarray miARN. Finalmente, via celular e análise de redes de dados microarray é explicado. Estas técnicas vão facilitar a comparação dos perfis de miRNA de três tipos de células diferentes.
